跑台训练对大鼠脑缺血后微血管生成及MT1-MMP表达影响

【摘要】:目的:通过观察跑台训练对脑缺血大鼠脑缺血周边区血管再生及MT1-MMP表达的情况,探讨跑台训练对大鼠脑缺血后微血管生成的影响及其可能机制。 方法:随机将42只成年雄性SD大鼠分为:假手术组(n=6),模型组(n=18)和运动组(n=18);采用改良Zea-Longa线栓法制备大鼠大脑中动脉栓塞(MiddleCerebral Artery Occlusion,MCAO)模型;在制备动物模型前,,所有大鼠先适应性跑台训练3天;模型制备成功后24小时对运动组大鼠进行跑台运动训练,采用改良的神经行为学评分方法(mNSS),分别在造模成功后4天、7天、14天对各模型组及运动组大鼠的神经功能恢复情况进行评价。分别于术后4天、7天、14天将大鼠断头取脑,用MVD法分析计数各组大鼠缺血脑组织周边区微血管数量;采用Western印迹法、免疫荧光组织化学法检测MT1-MMP的表达。 结果:与MCAO组大鼠相比,运动组大鼠脑缺血周边区的微血管数量及MT1-MMP表达水平明显增高,差异具有统计学意义(P0.05)。 结论: 跑台运动训练明显促进大鼠脑缺血周边区微血管新生,并能改善脑缺血后神经功能;其机制可能与MT1-MMP上调表达有关。
[Abstract]:Aim: to observe the effect of treadmill training on vascular regeneration and MT1-MMP expression in ischemic area of rats with cerebral ischemia, and to explore the effect of treadmill training on microangiogenesis after cerebral ischemia in rats and its possible mechanism.Methods: Forty-two adult male Sprague-Dawley rats were randomly divided into sham operation group (n = 6) and exercise group (n = 18). The middle cerebral artery occlusion (MCAO) model was established by modified Zea-Longa thread embolization method.All the rats were trained with adaptive treadmill for 3 days, and the rats in the exercise group were trained at 24 hours after the establishment of the model.A modified neurobehavioral scoring method was used to evaluate the recovery of neural function in rats of each model group and exercise group at 4 days, 7 days and 14 days after the establishment of the model.After 4 days, 7 days and 14 days after operation, the rat head was cut off and the number of microvessels in the ischemic brain tissue was counted by MVD method, and the expression of MT1-MMP was detected by Western blotting and immunofluorescence histochemistry.Results: compared with the MCAO group, the number of microvessels and the expression of MT1-MMP in the peripheral areas of cerebral ischemia in the exercise group were significantly higher than those in the MCAO group, and the difference was statistically significant (P 0.05).Conclusion:The treadmill training can obviously promote the microangiogenesis and improve the neural function in the cerebral ischemic area of rats, and its mechanism may be related to the up-regulation of MT1-MMP expression.
