MTT法体外检测脑胶质瘤对常用化疗药物敏感性研究

【摘要】:目的:运用MTT法测定脑胶质细胞瘤体外对化疗药物的敏感性,,为临床个性化化疗方案的设计提供参考。方法:取术中快速冰冻病理检查及术后常规病理检查均证实为胶质瘤并成功原代培养22例标本,与肿瘤化疗药多柔比星(Doxorubicin, ADM)、卩比柔比星(Pirarubicin,THP)、卡莫司汀(Carmustine, BCNU)、替尼泊苷(Teniposide, Vm-26)、顺铀(cis-Dichlorodiamineplatinum, DDP)的临床常用浓度(按血團峰值浓度(PPC)配制)作用72小时,MTT法检测不同肿瘤个体对不同化疗药物及其相互两两配对组合的敏感和耐药情况。结果:22例临床脑胶质瘤标本对5种化疗药物单独用药的敏感性从高到低依次为Vm-26 BCNU THPADM DDP,22例临床脑胶质瘤标本对5化疗药物两两配对的敏感性从高到低依次为THP+Vm-26 BCNU+Vm-26 ADM+Vm-26 Vm-26+DDP THP+BCNU ADM+BCNU BCNU+DDP THP+DDP ADM+DDP THP+ADM。年龄、性别对大多数药物敏感率无影响,肿瘤分级只对部分药物敏感率有影响。结论:MTT法体外药敏实验是一种简便、经济、准确的药敏实验,对于指导临床合理地制定个体化化疗方案有着重要的作用,5种常用的抗肿瘤药物均有耐药的情况,敏感性高低不一,单独用药则以Vm-26效果最好,而肿瘤细胞耐药性最强的则是DDP。但经过将化疗药物两两配对联合用药能明显提高杀瘤效果,其中以Vm-26+THP的组合效果最为明显,而将THP+ADM进行实验并不能提高药物敏感性,和单独用药无明显差别。
[Abstract]:Objective: to determine the sensitivity of glioma of brain to chemotherapeutic drugs by MTT method in order to provide reference for the design of individualized chemotherapy regimen. Methods: 22 cases of glioma were confirmed by intraoperative frozen pathological examination and routine pathological examination after operation and successfully cultured in primary culture. A 72 hour assay was used to detect the clinical concentrations of docorubicin, ADMN, pirarubicinine, Carmustine, BCNU, Teniposide, Vm-26N, cis-Dichodiamplatine inum (DDPum) in different tumor individuals by 72 hours of treatment with Doxorubicin, ADMN, pirarubicinine, Carmostin, Carmustine, Teniposide, Vm-26N, cis-Dichodiamplatineinum (DDPumn), which were prepared according to the peak blood mass concentration (PPCMTT). Sensitivity and resistance of chemotherapeutic agents and their paired combinations. Results the sensitivity of 22 brain glioma specimens to 5 chemotherapeutic drugs was from high to low. The sensitivity of 22 cases of glioma patients with clinical brain glioma was from high to low, from high to low to that of THP Vm-26. BCNU Vm-26 ADM Vm-26 Vm-26 DDP THP BCNU ADM BCNU BCNU DDP THP DDP ADM DDP THP ADM. Age and sex had no effect on most drug sensitivity, but tumor grading had no effect on drug sensitivity. Conclusion the in vitro drug sensitivity test with the method of: MTT is a simple, economical and accurate drug sensitivity test. It has an important role in guiding clinical rational formulation of individualized chemotherapy regimen. All of the five commonly used antitumor drugs are resistant to drugs. The sensitivity of Vm-26 was the best, and the drug resistance of tumor cells was the strongest. But the combination of two and two chemotherapeutic drugs can obviously improve the effect of tumor killing, among which the combination of Vm-26 THP is the most obvious, but the experiment of THP ADM can not improve the drug sensitivity, and there is no significant difference between the two groups.
