鼠脑胶质瘤瘤周水肿组织MR灌注成像与AQP1的相关性研究

【摘要】:目的:研究Wistar大鼠脑胶质瘤瘤周水肿组织的磁共振灌注成像(MR-PWI)的特征,并与免疫组化水通道蛋白-1(AQPl)对照,探讨胶质瘤瘤周水肿组织AQP1的表达与局部脑血容量(rCBV)的相关性。 方法:通过立体定向技术将C6细胞株种植于35只Wistar大鼠右侧大脑尾状核区,种植约3-4周后所有存活大鼠行常规MRI、PWI及T1WI增强检查。借助Function Tool软件对PWI数据后处理,选取瘤周水肿组织及相应对侧正常脑组织等大的3个感兴趣区,获得相应部位rCBV值。MRI扫描结束后立即处死大鼠,取全脑固定并做瘤周水肿区脑组织的连续切片,行相应部位的HE染色及AQP1免疫组化染色。利用Pearson相关分析法确定rCBV值与AQP1阳性表达IOD值之间的关系。 结果:接种C6细胞株3-4周后,大鼠胶质瘤瘤周水肿组织3个等大的感兴趣区rCBV平均值为(244.34±45.96),与对侧正常脑组织rCBV平均值(205.18±30.46)相比明显升高,差异具有统计学意义(P0.05)。瘤周水肿组织AQP1表达的IOD平均值为(76595.35±1116.98),瘤周水肿组织的rCBV值与AQP1阳性表达IOD值间均存在正相关(r=0.51,P0.05)。 结论:Wistar大鼠C6脑胶质瘤瘤周水肿组织的rCBV值较对侧脑组织升高,且可以反映AQP1的表达水平。
[Abstract]:Objective: to study the characteristics of Mr perfusion imaging (MR-PWI) in the peritumoral edema of gliomas in Wistar rats, and to investigate the correlation between the expression of AQP1 and regional cerebral blood volume (CBV) in glioma peri-tumorous edema tissues, as compared with immunohistochemical aquaporin-1 aquaporin (AQP). Methods: C6 cell lines were implanted in the right caudate nucleus of the brain of 35 Wistar rats by stereotactic technique. After implantation for about 3-4 weeks, all the surviving rats were examined with conventional MRI PWI and T1WI enhancement. Using Function Tool software to post-process the PWI data, three large regions of interest, such as the edema tissue around the tumor and the corresponding contralateral normal brain tissue, were selected, and the corresponding rCBV value was obtained. The rats were killed immediately after the scanning. The whole brain was fixed and serial sections of the brain tissue in the peritumoral edema area were made. The HE staining and AQP1 immunohistochemical staining were performed in the corresponding areas. Pearson correlation analysis was used to determine the relationship between rCBV value and IOD value of AQP1 positive expression. Results: after 3 to 4 weeks of C6 cell line inoculation, the average rCBV of 3 isometric regions of interest was 244.34 卤45.96, which was significantly higher than that of the contralateral normal brain tissue (205.18 卤30.46), and the difference was statistically significant (P 0.05). The average IOD expression of AQP1 in peritumoral edema tissue was 76595.35 卤1116.98, and there was a positive correlation between rCBV value and AQP1 positive expression IOD value in peritumoral edema tissue. Conclusion the rCBV value of brain edema tissue around C6 glioma is higher than that of contralateral brain tissue, and can reflect the expression level of AQP1.
