MicroRNA218在胶质瘤中的表达及对胶质瘤细胞迁移与侵袭的影响

【摘要】:背景和目的: 中枢神经系统肿瘤中脑肿瘤占90%,其中胶质瘤是最常见的脑肿瘤。胶质瘤细胞具有高度的侵袭、迁移及增殖能力。然而,促进肿瘤进展和复发的分子机制仍不确定。因此,治疗肿瘤的方法和药物非常有限,且多数患者的中位生存率不足1年,胶质母细胞瘤患者的临床生存率则更低。因而,探索胶质瘤细胞迁移及侵袭的分子机制是极有临床意义的,可以促进更有效的治疗手段或药物的研发。MicroRNAs是一类18至25个核苷酸长度的非编码单链的小RNAs,通过调控特定的有活性的mRNA靶点,,可在恶性肿瘤的进展过程起重要作用。超过半数的人类基因由microRNA调控转录抑制和(或)mRNA降解来调控。MicroRNAs也参与肿瘤细胞的凋亡、增殖、迁移及侵袭的调控。研究表明,microRNA与肿瘤的发生有密切关系,并可分为致癌microRNA和抑癌microRNA。MicroRNA218为抑癌microRNA。近年来,MicroRNA218在一些肿瘤中已广泛被研究,研究表明其在脑胶质瘤细胞中显著表达下调。然而,MicroRNA218对胶质瘤细胞的迁移和侵袭的生物学影响知之甚少。本研究旨在探讨microRNA218在脑胶质瘤组织样本中的表达水平与病理分级的相关性,以及microRNA218的生物学功能。 方法: (1)收集经病理确诊的不同级别胶质瘤的新鲜组织样本38例,用液氮罐转运至㧟80℃冰箱,用于总RNA的提取。 (2)应用定量RT-PCR检测miRNA218在各脑胶质瘤组织样本中的表达水平,并分析其与胶质瘤病理分级的相关性。 (3)应用脂质体2000将pcDNA3.1-microRNA218质粒转染胶质瘤细胞U87、U251。 (4)使用细胞计数试剂盒-8(CCK-8)细胞计数法检测microRNA218对胶质瘤细胞U87、U251存活率的影响。 (5)应用Transwell侵袭实验检测microRNA218对胶质瘤细胞U87的侵袭能力的影响。 (6)应用细胞划痕实验测定microRNA218对胶质瘤细胞U87的迁移能力的影响。 结果: (1)MicroRNA218在WHOI-II胶质瘤患者中的表达水平明显高于WHOIII-IV患者,且microRNA218的表达水平与脑胶质瘤病理分级呈负相关。 (2)MicroRNA218抑制U87MG和U251MG细胞增殖能力。 (3)MicroRNA218体外抑制胶质瘤细胞U87迁移与侵袭能力。结论: (1)MicroRNA218表达水平与病理分级呈负相关。 (2)MicroRNA218具有抑制胶质瘤迁移与侵袭的作用。-
[Abstract]:Background and objective: brain tumors account for 90% of central nervous system tumors, of which glioma is the most common brain tumor. Glioma cells have high ability of invasion, migration and proliferation. However, the molecular mechanism for tumor progression and recurrence remains uncertain. Therefore, the treatment of tumors and drugs are very limited, and the median survival rate of most patients is less than 1 year, and the clinical survival rate of glioblastoma patients is even lower. Therefore, it is of great clinical significance to explore the molecular mechanism of migration and invasion of glioma cells. MicroRNAs are a class of non-coding single-stranded RNs with 18 to 25 nucleotides, which play an important role in the progression of malignant tumors by regulating specific active mRNA targets. More than half of human genes are regulated by microRNA to regulate transcription inhibition and / or mRNA degradation. MicroRNAs are also involved in the regulation of apoptosis, proliferation, migration and invasion of tumor cells. The results showed that microRNAs were closely related to tumorigenesis and could be divided into carcinogenic microRNA and tumor suppressor microRNA.MicroRNA218 as tumor suppressor microRNA.
