慢性脑缺血大鼠脑组织NogoA、NgR1蛋白表达及山楂叶总黄酮的干预作用

【摘要】:目的探讨山楂叶总黄酮对慢性脑缺血大鼠脑组织轴突过度再生抑制因子(Nogo)A及其受体NgR1表达的影响。方法健康雄性SD大鼠,随机分为假手术组、慢性脑缺血模型组、山楂叶总黄酮组和银杏叶片组。采用双侧颈总动脉永久性双重结扎法建立慢性脑缺血模型。HE染色法观察神经元密度;Western印迹和免疫组化法检测缺血脑组织Nogo A和NgR1蛋白的表达。结果与假手术组海马神经细胞比较,模型组大鼠CA1区神经细胞排列混乱,神经元细胞间隙增大,部分细胞溶解消失,细胞核固缩、浓染,正常神经元数量较假手术组减少(P0.01),脑组织内Nogo A(P0.01)和NgR1(P0.05)蛋白表达升高;与模型组比较,山楂叶总黄酮组病变明显减轻,正常的神经细胞排列规则,结构较完整,神经元密度增加(P0.05),Nogo A(P0.01)和NgR1(P0.05)蛋白表达降低。结论山楂叶总黄酮具有下调Nogo A和NgR1蛋白表达,促进脑神经元轴突再生,保护慢性脑缺血大鼠神经的功能。
[Abstract]:Objective to investigate the effects of total flavonoids in Hawthorn leaves on the expression of (Nogo) A and its receptor NgR1 in brain tissue of rats with chronic cerebral ischemia. Methods healthy male Sprague-Dawley rats were randomly divided into sham-operation group, chronic cerebral ischemia model group, Hawthorn leaf flavonoid group and ginkgo leaf group. The model of chronic cerebral ischemia was established by permanent double ligation of bilateral common carotid artery. The expression of Nogo A and NgR1 in ischemic brain tissue was detected by Western blot and immunohistochemistry. Results compared with the sham operation group, the nerve cells in the CA1 region of the model group were disordered, the gap between the neurons increased, some cells dissolved and disappeared, and the nucleus became pyknosis and staining. Compared with the model group, the number of normal neurons decreased (P0.01), and the expression of Nogo A (P0.01 and NgR1 (P0.05) protein increased in brain tissue. The expression of Nogo A (P0.01) and NgR1 (P0.05) decreased. Conclusion Total flavonoids in Hawthorn leaves can down-regulate the expression of Nogo A and NgR1 proteins, promote axonal regeneration of brain neurons, and protect the nerves of rats with chronic cerebral ischemia.
