Nav1.8离子通道在SNI大鼠背根神经节及外周神经的表达变化

【摘要】:目的:通过观察保留性神经损伤(SNI)大鼠模型中Nav1.8在受损背根神经节(dorsal root ganglion,DRG)、外周轴突以及表皮神经纤维中的表达变化,探讨Nav1.8蛋白在部分神经损伤所致神经病理性疼痛中的可能机制。方法:选择清洁级SD大鼠12只,随机分为两组:保留性神经损伤组(SNI组)与假手术组(Sham组);造模时分别在SNI组和Sham组大鼠腓肠神经鞘内注射荧光金(FG),造模当天和造模后第7天与第14天测量两组大鼠机械刺激伤害感受阈值(MWT)。术后14天处死SNI组和Sham组大鼠,取L5 DRG、腓肠神经、胫神经、腓总神经组织,以及腓肠神经和胫神经支配区域皮肤组织。采用免疫荧光技术检测两组大鼠Nav1.8在DRG FG阳性(FG+)区域和阴性(FG-)区域,腓肠神经、胫神经、腓总神经,以及腓肠神经和胫神经支配皮肤区域的表达变化。结果:L5 DRG中FG+区域与FG-区域呈区域分布。术后第7天和第14天,SNI组大鼠的MWT值较Sham组明显降低(P0.01)。Nav1.8在SNI组大鼠腓肠神经、胫神经、腓总神经以及腓肠神经支配皮肤区域的表达显著高于Sham组(P0.05),而Nav1.8在胫神经支配皮肤区域的表达明显低于Sham组(P0.05)。Nav1.8在SNI组大鼠DRG FG+和FG-区域的表达高于Sham组(P0.05),而在SNI组内,DRG FG+区域的Nav1.8表达显著高于FG-区域(P0.05)。结论:DRG中腓肠神经投射区域与胫神经、腓总神经投射区域呈解剖分离状态。Nav1.8在DRG、未损伤神经及支配皮肤表达的增高可能是部份神经损伤引起慢性神经病理性疼痛的致病因素之一。
[Abstract]:Objective: to observe the expression of Nav1.8 in the injured dorsal root ganglion (dorsal root ganglion,DRG), peripheral axons and epidermal nerve fibers in (SNI) rats. To explore the possible mechanism of Nav1.8 protein in neuropathic pain caused by partial nerve injury. Methods: twelve clean grade SD rats were selected. Two groups were randomly divided into two groups: the SNI group (SNI group) and the sham operation group (Sham group), and the rats in SNI group and Sham group were given intrathecal injection of fluorescent gold (FG), on the same day and on the 7th and 14th days after modeling. (MWT). Of nociceptive threshold of mechanical stimulation in rats On the 14th day after operation, the rats in SNI group and Sham group were killed. The sural nerve, tibial nerve, common peroneal nerve, sural nerve and tibial nerve innervated area skin tissue of L 5 DRG, were taken out. Immunofluorescence technique was used to detect the expression of Nav1.8 in the areas of DRG FG positive (FG) and FG-, sural nerve, tibial nerve, common peroneal nerve, sural nerve and tibial nerve innervated skin. Results: the region of FG and FG- were distributed in L 5 DRG. On the 7th and 14th day after operation, the MWT value of SNI group was significantly lower than that of Sham group (P0.01). Nav1.8 was observed in sural nerve and tibial nerve in SNI group. The expression of common peroneal nerve and sural nerve innervated skin area was significantly higher than that of Sham group (P0.05), while the expression of Nav1.8 in tibial nerve innervation skin area was significantly lower than that in Sham group (P0.05). The expression of Nav1.8 in DRG FG and FG- region of SNI group was higher than that of Sham group (P0.05), while in SNI group, the expression of Nav1.8 was higher in SNI group than in Sham group (P0.05). The expression of Nav1.8 in, DRG FG was significantly higher than that in FG- (P0.05). Conclusion: the area of sural nerve projection and tibial nerve in DRG, The increased expression of Nav1.8 in the uninjured and innervated skin of DRG, may be one of the pathogenic factors of chronic neuropathic pain caused by partial nerve injury.
