亚低温对大鼠脑缺血再灌注海马nestin、caspase-3阳性细胞表达的影响

【摘要】:目的:本研究采用Longa线栓法建立大鼠脑缺血再灌注模型,观察比较亚低温治疗组与常温组大鼠神经功能缺损评分、脑梗死体积及海马区nestin、caspase-3阳性细胞表达情况,目的是探索脑缺血后亚低温治疗的神经保护作用、亚低温治疗对神经再生与细胞凋亡的影响及可能机制,为临床缺血性脑血管病的亚低温治疗提供理论依据。 方法:将144只SD大鼠随机分为三组:假手术组、缺血再灌注+常温组、缺血再灌注+亚低温组。常温组、低温治疗组据缺血后再灌注时间不同各分为1天、3天、7天、14天四个亚组。采用Longa线栓法建立大鼠脑缺血再灌注模型,亚低温组于脑缺血模型完成后即刻予4h的全身亚低温治疗,亚低温治疗期间维持大鼠直肠温度在33-35℃之间,常温组及假手术组大鼠直肠温度维持在36.8~37.2℃之间。Longa评分法评估造模前、缺血再灌注各时间点大鼠的神经功能缺损程度,TTC染色法测量脑梗死体积,免疫组化法检测海马区nestin阳性细胞及caspase-3阳性细胞表达的情况。 结果: 1.神经功能缺损评分:假手术组、常温组、低温组三组大鼠术前Longa评分均为0分,无统计学差异。术后假手术组大鼠无神经功能缺损症状。常温组、低温组大鼠再灌注后1天、3天、7天、14天四个时间点神经功能评分数值呈逐渐降低趋势。再灌注后1天低温组大鼠神经功能缺损评分与常温组比较无显著差异(P0.05),其他三个时间点低温组的神经功能评分均较常温组降低(P0.05)。 2.大鼠脑梗死体积比较:假手术组术后无梗死。常温组、低温组均表现为再灌注后1天梗死体积开始增加,再灌注后3天梗死最明显,随后逐渐下降。再灌注后1天、3天、7天、14天低温组大鼠脑梗死体积与常温组比较均明显减小(P0.05)。 3.海马nestin阳性细胞表达:假手术组大鼠海马可见少量nestin阳性细胞表达。常温组、低温组海马nestin阳性细胞数随着再灌注时间的延长呈逐渐增加趋势,再灌注后3天、7天阳性细胞表达处于较高水平。再灌注后1天、3天低温组nestin阳性细胞数目均较常温组增多(P0.05),但再灌注后7天、14天低温组与常温组比较阳性细胞表达无明显差异(P0.05)。 4.海马caspase-3阳性细胞表达:假手术组可见少量阳性细胞表达。常温组、低温组再灌注后1天时caspase-3阳性细胞表达均处于较高水平,再灌注后3天开始阳性细胞表达逐渐下降。再灌注后1天、3天、7天低温组阳性细胞数目均较常温组降低(P0.05),但再灌注后14天两组阳性细胞数目比较无明显差异(P0.05)。 结论: 1.亚低温(33-35℃)治疗能够改善SD大鼠脑缺血再灌注后神经功能缺损症状、降低脑梗死体积、促进海马区nestin阳性细胞的表达及抑制caspase-3阳性细胞的表达。 2.亚低温(33~35℃)治疗SD大鼠脑缺血再灌注损伤,具有良好的神经保护作用,这可能与亚低温治疗促进海马区nestin阳性细胞表达、抑制caspase-3阳性细胞表达有关。
[Abstract]:Objective: to establish a rat model of cerebral ischemia-reperfusion by Longa thread embolization, and to observe and compare the neurological deficit score, cerebral infarct volume and the expression of nestin,caspase-3 positive cells in hippocampus between mild hypothermia group and normothermic group. Objective to explore the neuroprotective effect of mild hypothermia therapy after cerebral ischemia, the effect of mild hypothermia therapy on nerve regeneration and apoptosis and its possible mechanism, and to provide theoretical basis for mild hypothermia therapy of clinical ischemic cerebrovascular disease.
