大动脉粥样硬化型脑梗死患者TLR4基因多态性观察

【摘要】:目的观察中国胶东半岛地区大动脉粥样硬化(LAA)型脑梗死汉族患者TLR4基因的rs1927911、rs1927914、rs2149356位点单核苷酸多态性情况。方法将LAA型脑梗死患者105例纳入病例组,选择同期健康体检者125例纳入对照组。采用PCR-RFLP法或直接测序法观察两组白细胞TLR4基因rs1927911、rs1927914、rs2149356位点的单核苷酸多态性情况。结果两组各多态性位点的基因型和等位基因频率均符合Hardy-Weinberg遗传平衡定律(P0.05)。病例组TLR4基因rs1927911位点CC、TT基因型频率低于对照组,CT基因型频率高于对照组(P均0.05)。病例组rs2149356位点CC、AC基因型频率低于对照组,AA基因型频率高于对照组,A等位基因频率高于对照组(P均0.05)。结论中国胶东半岛地区LAA型脑梗死汉族患者白细胞TLR4基因rs1927911、rs2149356位点多态性可能与LAA型脑梗死的发病有关。
[Abstract]:Objective to observe the single nucleotide polymorphism (SNP) of TLR4 locus of TLR4 gene in patients with (LAA) type cerebral infarction in Jiaodong Peninsula. Methods 105 patients with LAA type cerebral infarction were included in the case group, and 125 healthy persons were included in the control group. Single nucleotide polymorphisms at rs1927911,rs1927914,rs2149356 site of TLR4 gene were observed by PCR-RFLP or direct sequencing. Results the genotypes and allelic frequencies of each polymorphism locus in the two groups were in accordance with the Hardy-Weinberg genetic balance law (P0.05). The frequency of CC,TT genotype at rs1927911 site of TLR4 gene was lower than that of control group, and the frequency of CT genotype was higher than that of control group (all P 0.05). The frequency of CC,AC genotype, AA genotype and A allele in rs2149356 locus were lower than those in control group (P < 0. 05). Conclusion the polymorphisms of TLR4 gene rs1927911,rs2149356 locus of leukocyte in Han nationality patients with LAA type cerebral infarction in Jiaodong Peninsula of China may be related to the pathogenesis of LAA type cerebral infarction.
