TRPV4在小鼠脑血管内皮细胞中的表达及对血管张力调节作用研究
[Abstract]:Objective to compare the expression of vanillin receptor subfamily (TRPV) and classical transient receptor potential subfamily (TRPC) in mouse brain, heart and liver microvascular endothelial cells. To explore the specific high expression subtypes of cerebral microvessels and elucidate its regulatory effect on cerebral artery vasodilation. Methods the microarray expression profile data in GEO database were excavated, the differential expression of each channel subgroup was analyzed, and then the expression of TRPV4,TRPP2 and TRPC1 in cerebral basilar artery was detected by immunohistochemistry. The effect of TRPV4 channel on the relaxation of basilar artery in mice was detected by vascular tension test. Results the expression profile data showed that TRPV4 channel was significantly overexpressed in cerebral microvascular endothelial cells, while TRPC3 channel was significantly lower in mouse brain, heart and liver microvascular endothelial cells. TRPV4, TRPV4, TRPP2 and TRPC1 were also significantly expressed in the endothelial and smooth muscle layers of basilar artery. TRPV4 channel agonist GSK1016790A could relax the contraction of basilar artery induced by phenylephrine in a concentration-dependent manner, and its relaxation effect could be blocked by TRPV4 channel blocker HC067047. Conclusion compared with cardiac and hepatic vascular endothelial cells, TRPV4 channel is significantly overexpressed in mouse cerebral vascular endothelial cells, which may play an important role in the regulation of cerebral vascular endothelial function.
