PAR2和TMEM16A在神经病理性疼痛模型大鼠背根神经节上的表达及意义

【摘要】:目的观察坐骨神经慢性压迫模型(CCI)的大鼠背根神经节(DRG)神经元上PAR2和TMEM16A的表达,探讨其在神经病理性疼痛中发挥的作用。方法大鼠分为假手术组(Sham)和CCI组,两组分别检测热缩足反射潜伏期(TWL);采用免疫荧光和Western blot检测大鼠DRG神经元上PAR2和TMEM16A表达。结果与Sham组比较,CCI组术前TWL差异无统计学意义,术后各时间点均明显降低且差异具有统计学意义(P0.01)。免疫荧光结果显示:PAR2与TMEM16A在大鼠DRG神经元上共存。Western blot结果显示:与Sham组相比,CCI组术后7、14 d的PAR2和TMEM16A蛋白表达均出现明显增加(P0.01),且CCI组14 d高于7 d的PAR2和TMEM16A蛋白表达(P0.05)。结论神经病理性疼痛大鼠DRG神经元上PAR2和TMEM16A蛋白表达水平较Sham组显著升高,这些蛋白表达水平升高可能是导致大鼠神经病理性疼痛的原因之一。
[Abstract]:Objective to observe the expression of PAR2 and TMEM16A in (DRG) neurons of dorsal root ganglion of rats with chronic compression of (CCI), and to explore the role of PAR2 and TMEM16A in neuropathic pain. Methods the rats were divided into (Sham) group and CCI group. The latent period of thermal contractile foot reflex (TWL); was detected by immunofluorescence and Western blot. The expression of PAR2 and TMEM16A in DRG neurons was detected by immunofluorescence and Western blot respectively. Results compared with Sham group, there was no significant difference in TWL in CCI group before operation, but significantly decreased at each time point after operation (P 0.01). The results of immunofluorescence showed that PAR2 and TMEM16A coexisted on DRG neurons of rats. Western blot results showed that the expression of PAR2 and TMEM16A protein in CCI group was significantly higher than that in Sham group on the 14th day after operation (P01), and the expression of PAR2 and TMEM16A protein in CCI group was higher than that on 7th day (P 0.05). Conclusion the expression levels of PAR2 and TMEM16A proteins in DRG neurons of rats with neuropathic pain are significantly higher than those in Sham group. The increased expression of these proteins may be one of the causes of neuropathic pain in rats.
