BDNF/TrkB通路和SIRT1参与硫化氢拮抗同型半胱氨酸诱导大鼠海马神经元损伤

【研究背景与目的】同型半胱氨酸(Homocysteine, Hcy)诱导神经细胞死亡在阿尔茨海默病(Alzheimer’s disease, AD)的病理进展中具有重要作用。硫化氢(Hydrogen sulfide,H2S)是一种内源性神经保护剂,具有抗氧化、抗凋亡作用。本课题组的以往研究发现H2S能够拮抗Hcy的神经细胞毒性,但其机制尚未阐明。脑源性神经营养因子(Brain derived neurophic factor, BDNF)是由神经元分泌的,在神经细胞的分化、存活、发育中起到重要作用。作为Sirtuins家庭中的一员,沉默信息调节因子(Silent information regulator, SIRT1)在大脑、小脑及海马中均有表达,对于调节细胞的新陈代谢、衰老等起到重要作用。因而,我们将从BDNF和SIRT1的角度探讨H2S拮抗Hcy神经毒性的机制。为此,本实验以侧脑室注射Hcy的SD大鼠为Hcy神经毒性的动物模型,探讨H2S对Hcy损伤海马神经元的拮抗作用以及BDNF/TrkB通路和SIRT1对H2S抗Hcy损伤海马神经元的介导作用。【方法】Tunel (Termi...
