载红霉素衍生物LY267108骨水泥洗提特性的实验研究

【摘要】:目的:探讨红霉素衍生物LY267108加入骨水泥后,其体外的释放规律。方法:实验共分为5组,A1-A5为在40克骨水泥中分别加入0.5g、1g、1.5g、2g、2.5gLY267108制成模型,放入模拟人工体液进行浸提,并于设定的时间点,从每个样本浸提液中,提取1m1浸提液,储存于-20℃的冰箱中,实验完成后用高效液相色谱法测定不同时间点药物的释放量。结果:在第1周内(1d-7d),5组药物均呈爆发性释放,各组间相同时段释放比率差异无统计学意义(P0.05),第7天,各组释放比率约为11%左右;自第2周至第8周间(8d-56d)药物的释放速度明显减慢,缓慢释放期内在各组LY267108累计释放速度差异有统计学意义(P0.05),且比较各组累计释放速率A1组A2组A5组A3组A4组。结论:在第1周(1d-7d),LY267108呈爆发性释放,且各组相同时段的释放百分率相似,自第2周至第8周(8d-56d)LY267108的释放速度明显减慢;在40g骨水泥中加入2.0g LY267108,LY267108更趋于稳定、持久的从骨水泥中释放。
[Abstract]:Objective: To investigate the effect of erythromycin derivatives LY267108 into bone cement after release in vitro. Methods: mice were divided into 5 groups, A1-A5 0.5g, were added in 40 grams of bone cement in 1g, 1.5g, 2G, 2.5gLY267108 made into model, extracting the simulated body fluid, and to set time points, from each sample extract, extract 1m1 extract, refrigerator stored in -20 DEG C, after completion of the experiment was determined by HPLC at different time point of drug release. Results: in first weeks (1d-7d), the 5 groups showed the same time the explosive release, release ratio between groups there was no statistically significant difference (P0.05), seventh days, each release ratio is about 11%; from second to eighth weeks (8d-56d) between the drug release rate slowed down, slow release period of each LY267108 cumulative release rate was statistically significant difference (P0.05), and compared the The cumulative release rate of A1 group A2 group A5 group A3 group A4 group. Conclusion: at first weeks (1d-7d), LY267108 explosive release, the release percentage and similar groups of the same period, from second to eighth weeks (8d-56d) LY267108 release rate slowed down; adding 2.0g LY267108 in 40g bone cement, LY267108 more stable, sustained release from bone cement.
