成年裸鼠与野生型小鼠皮肤形态结构及表皮干细胞标志物分布、表达量的比较研究

【摘要】:目的比较成年裸鼠与野生型小鼠皮肤形态结构及表皮干细胞标志物p63、角蛋白19和β1整合素的分布和表达量的差异,为后续的皮肤组织工程学研究提供客观的参照。方法分别取8只成年裸鼠与野生型小鼠背部皮肤,制作石蜡切片,进行HE染色,光镜下观察皮肤表皮、真皮和皮肤附属器的形态结构;免疫组织化学法检测p63、角蛋白19和β1整合素在两种鼠皮肤的分布和表达量。结果 HE染色低倍镜下裸鼠表皮较野生型小鼠厚,高倍镜下裸鼠表皮由3~4层上皮细胞组成,而野生型小鼠表皮由1~2层上皮细胞组成。裸鼠表皮基底层细胞呈立方形或者矮柱形,细胞核深染,呈卵圆形,居中,野生型小鼠表皮基底层细胞呈立方形,细胞核圆形深染。裸鼠表皮有明显的颗粒层,而野生型小鼠表皮则无。裸鼠皮脂腺没有野生型小鼠皮脂腺发达;免疫组织化学显示p63表达于裸鼠和野生型小鼠毛囊和表皮基底层细胞的胞核中,角蛋白19表达于裸鼠和野生型小鼠皮肤毛囊和基底层细胞中,β1整合素表达于裸鼠皮肤毛囊和表皮颗粒层细胞中及野生型小鼠毛囊中;裸鼠p63、β1整合素和角蛋白19的表达量少于野生型小鼠,差异有统计学意义(P0.01)。结论成年裸鼠与野生型小鼠皮肤形态结构及表皮干细胞标志物的分布和表达量存在差异,可以作为区别裸鼠与野生型小鼠皮肤的参考。
[Abstract]:Objective to compare the distribution and expression of epidermal stem cell markers p63, keratin 19 and 尾 1 integrin in adult nude mice and wild type mice. Methods the back skin of 8 adult nude mice and wild-type mice were taken to make paraffin sections and stained with HE. The skin epidermis was observed under light microscope. The distribution and expression of p63, keratin 19 and 尾 1 integrin in the skin of mice were detected by immunohistochemistry. Results the epidermis of nude mice was thicker than that of wild-type mice under HE staining. The epidermis of nude mice was composed of three or four layers of epithelial cells under high power microscope, while the epidermis of wild type mice was composed of 1 layer and 2 layers of epithelial cells. The basal layer of epidermis of nude mice was square or short columnar, and the nucleus was deep stained, oval and intermediate. The basal layer cells of wild mouse epidermis were square, and the nuclei were round and deep stained. The epidermis of nude mice had obvious granular layer, but the epidermis of wild type mice was not. The sebaceous glands of nude mice were not as developed as those of wild type mice. Immunohistochemical staining showed that p63 was expressed in the nuclei of hair follicles and epidermal basal layer cells in nude mice and wild type mice. Keratin 19 was expressed in hair follicles and basal layer cells of nude mice and wild type mice, and 尾 1 integrin was expressed in hair follicles and granular layer cells of skin of nude mice and in hair follicles of wild-type mice. The expression of p63, 尾 1 integrin and keratin 19 in nude mice was lower than that in wild type mice, and the difference was statistically significant (P 0.01). Conclusion there are differences in skin morphology, distribution and expression of epidermal stem cell markers between adult nude mice and wild type mice. It can be used as a reference to distinguish the skin between nude mice and wild mice.
