海藻酸盐对CS-GP可注射水凝胶温敏性能的影响

【摘要】:针对干眼症临床治疗对泪点栓产品的研发需求,文中利用壳聚糖(CS)与β-甘油磷酸钠(GP)可在37℃发生温敏相变形成凝胶的特性,采用生物相容性优良的海藻酸钠(SA)为共混材料,改进CS-GP凝胶存在温敏相变时间较长、机械强度差、易于溶胀和降解、热稳定性差等问题,并采用扫描电子显微测试、红外光谱分析、热重分析、压缩弹性模量和流变性能分析等方法对复合凝胶性能进行了表征。结果表明:制备的复合溶胶在37℃环境可快速相变形成具有多孔网络结构且强度适宜的凝胶,可注射性良好。当CS质量浓度为25 mg/mL,GP质量浓度为600 mg/mL,V(CS)∶V(GP)=4.5∶1时,添加40 mg/mL SA可将CS-GP-SA复合凝胶的相变时间调控为10 s以内,压缩弹性模量为67 k Pa,损耗模量G″和储能模量G'值比为0.003 1,平衡溶胀率为5%,置于模拟泪液中48 d时的质量残留率为61%。凝胶性能可以满足泪道栓塞的应用需求。
[Abstract]:According to the research needs of clinical treatment of dry eye of punctal plug products, using the shell chitosan (CS) and sodium glycerophosphate (GP) can occur at 37 DEG C transition temperature sensitive gel forming properties, with excellent biocompatibility of alginate (SA) blend materials, improved CS-GP gel are temperature sensitive phase transition a long time, poor mechanical strength, easy to swelling and degradation problems, poor thermal stability, and using scanning electron microscopy, infrared spectroscopy, thermogravimetric analysis, compressive elastic modulus and rheological analysis were used to characterize the composite gel performance. The results showed that the preparation of the sol in 37 DEG C can be fast phase formation with porous network structure and the appropriate intensity of gel, injectable good. When CS concentration was 25 mg/mL, GP concentration was 600 mg/mL, V (CS): V (GP) =4.5: 1, add 40 mg/mL SA can be CS-GP-SA composite gel The phase transition time is controlled within 10 s, the compression modulus is 67 K Pa, the loss modulus G and storage modulus G'ratio is 0.0031, the equilibrium swelling rate is 5%, and the mass remaining rate in the simulated tear 48 D is 61%. gel, which can meet the application requirement of lacrimal duct embolization.
