三维钛网支架材料对犬牙周膜成纤维细胞生物学行为的影响

【摘要】:目的研究三维钛网支架(TW)材料对犬牙周膜成纤维附着、增殖、分化的影响。方法通过酶消化组织块法原代培养犬牙周膜成纤维细胞(PDLFs),传代后与TW材料接触培养,噻唑蓝(MTT)法比较实验组三维钛网培养与对照组二维培养对种子细胞的促增殖作用,通过提取测量上清液碱性磷酸酶含量比较实验组三维钛网培养与对照组传统矿化诱导液培养对种子细胞的促分化作用。结果酶消化组织块法可以成功获得用于实验的犬PDLFs,且细胞能有效的与TW附着,实验组的促增殖和分化作用要显著优于对照组。结论 TW材料能有效促进牙PDLFs的附着、增殖、分化,在牙周组织再生工程有良好的应用前景。
[Abstract]:Objective to study the effect of three dimensional titanium mesh scaffold (TW) on the attachment, proliferation and differentiation of canine periodontic membrane.Methods using enzyme digestion tissue block method, the primary cultured fibroblasts of perodontoid membrane were cultured in contact with the TW material. The proliferation of seed cells in the experimental group was compared with that in the control group by the method of MTT- (thiazolyl blue), three dimensional titanium mesh culture and the control group.By extracting and measuring the content of alkaline phosphatase in supernatant, the effects of three dimensional titanium mesh culture on the differentiation of seed cells in the experimental group and the traditional mineralized inducer culture in the control group were compared.Results PDLFs could be successfully obtained by enzyme digestion tissue block method, and the cells could attach to TW effectively. The effect of promoting proliferation and differentiation in the experimental group was significantly better than that in the control group.Conclusion TW can effectively promote the attachment, proliferation and differentiation of tooth PDLFs, and has a good application prospect in periodontal tissue regeneration engineering.
