含有磷酰胆碱基聚氨酯的合成及其结构—性能研究

【摘要】:新型抗凝血材料的合成和性能研究具有重大的应用价值和科学意义,是目前生物医用材料的热点研究领域之一。其中,在高分子材料上修饰磷酰胆碱结构的物质是有效提高材料抗凝血性的重要途径之一。磷酰胆碱结构修饰的高分子材料主要有两种类型,一类是磷酰胆碱修饰在高聚物的侧链上,另一类是磷酰胆碱结构在主链上。磷酰胆碱有效基团在主链上的聚合物由于具有与生物大分子更相似的聚电解质链结构和自组装形式,一直是人们关注的焦点,作为药物载体和植入材料在生物医药领域具有广泛的应用前景。在本论文中,我们成功合成了主链上含有磷酰胆碱结构的聚氨酯(PCPU),并利用红外、核磁等手段对其化学结构进行了精确表征。我们对制备得到的聚氨酯-磷酰胆碱聚氨酯(PU-PCPU)的复合膜材料进行了力学性能(拉伸应力和杨氏模量)和生物学性能(血小板粘附和溶血)的评估,证明其有望用于抗凝血材料的实际临床应用。同时,我们利用介电谱表征了固体膜和膜-水体系的介电性能。固体膜的介电测试结果表明由于偶极离子的凝聚使得PCPU的软段刚性增强,成功解释了复合膜杨氏模量的增强的结构原因。而膜-水体系的介电测试提供了一种原位表征材料表面性能的新方法,定量描述了膜表面的界面动力学,并由此提出关于血液或生物相容性机理的新假想,即含有两性离子结构的亲水性表面具有良好生物(血液)相容性是基于其对外界电磁刺激的响应特征。
[Abstract]:The research on the synthesis and properties of new anticoagulant materials has great application value and scientific significance, and is one of the hot research fields of biomedical materials.Among them, modification of phosphatidylcholine structure on polymer materials is one of the important ways to improve anticoagulability.There are two main types of phosphorylcholine modified polymer, one is phosphorylcholine modified on the side chain of polymer, the other is phosphorylcholine structure on the main chain.Phosphorylcholine effective group polymers on the main chain have been the focus of attention due to their polyelectrolyte chain structure and self-assembly form, which are more similar to biological macromolecules.As a drug carrier and implant material, it has a wide application prospect in the field of biomedicine.In this thesis, we have successfully synthesized PCPU with phosphatidylcholine structure on the main chain and characterized its chemical structure by IR and NMR.The mechanical properties (tensile stress and Young's modulus) and biological properties (platelet adhesion and hemolysis) of the PU-PCPU composite membranes were evaluated.It is proved that it is expected to be used in clinical application of anticoagulant materials.At the same time, the dielectric properties of solid films and film-water systems were characterized by dielectric spectroscopy.The results of dielectric measurements show that the rigidity of the soft segment of PCPU is enhanced due to the condensation of dipole ions, which explains the structural reasons for the enhancement of the Young's modulus of the composite membrane.The dielectric measurement of membrane-water system provides a new method for in situ characterization of the surface properties of the materials. The interfacial kinetics of the membrane surface is described quantitatively, and a new hypothesis about the mechanism of blood or biocompatibility is proposed.The hydrophilic surface with amphoteric ion structure has good biocompatibility based on its response to external electromagnetic stimulation.
