人羊膜治疗家兔皮肤Ⅲ度烧伤的作用与抗炎机制研究

【摘要】:目的:探索人羊膜治疗家兔皮肤Ⅲ度烧伤的作用和抗炎机制。 方法:热铜棒烫伤致兔腹背部皮肤Ⅲ度烧伤。切痂后,左侧创面分别帖敷新鲜人羊膜、涂抹rhEGF凝胶,右侧创面仅涂生理盐水。观测烧伤创面愈合率和创面痊愈时间,并行创面组织病理学检查。ELISA法检测早期创面组织中IL-1、IL-6、IL-8、IL-10、IL-12和TNF-α含量。 结果:①治疗第3d、8d和18d,羊膜组烧伤创面愈合率分别为(35.78±6.75)%、(71.21±6.63)%和(96.65±1.42)%,rhEGF凝胶组则为(23.75±3.57)%、(63.33±4.18)%和(92.79±1.80)%,对照组愈合率为(20.96±5.61)%、54.22±9.74)%和(90.73±2.15)%,羊膜组创面愈合率均显著高于rhEGF组(P0.05)和对照组(P0.01);羊膜组、rhEGF组、对照组创面分别于18.40±1.26d、20.20±1.69d和22.13±1.25d痊愈,羊膜组的愈合时间较rhEGF组和对照组显著缩短(P0.05)。②治疗第3d,羊膜组创面组织红肿较rhEGF组和对照组明显减轻。病理学检查显示,羊膜组的组织水肿明显较rhEGF组和对照组轻,炎细胞浸润也最少。③治疗第3和8d,羊膜组创面组织中IL-1、IL-6、 IL-8、IL-12和TNF-a含量显著低于rhEGF组和对照组(P0.05或P0.01),IL-10含量明显高于rhEGF组和对照组(P0.05或P0.01)。 结论:人羊膜能促进家兔皮肤Ⅲ度烧伤的愈合,在创面愈合早期通过下调创面组织中IL-1、IL-6、IL-8、IL-12和TNF-α的含量,上调IL-10的含量而发挥明显抗炎作用,减轻创面炎症反应。
[Abstract]:Objective: to explore the effect and anti-inflammatory mechanism of human amniotic membrane in treating the third degree burn of rabbit skin.Methods: the third degree burn of rabbit's abdominal back skin was caused by scalding with hot copper rod.After escharectomy, the left wound was coated with fresh human amniotic membrane and rhEGF gel, while the right wound was only coated with normal saline.The wound healing rate and wound healing time were observed, and the contents of IL-10 IL-12 and TNF- 伪 in early wound tissues were detected by Elisa and histopathological examination.The wound healing time in the amniotic membrane group was significantly shorter than that in the rhEGF group and the control group on the 3rd day, and the redness of the wound tissue in the amniotic membrane group was significantly less than that in the rhEGF group and the control group.Histopathological examination showed that the tissue edema in amniotic membrane group was significantly lighter than that in rhEGF group and control group.On the 3rd and 8th day after the treatment, the contents of IL-1IL-6, IL-8 IL-12 and TNF-a in the amniotic membrane group were significantly lower than those in the rhEGF group and the control group (P0.05 or P0.01). The contents of IL-10 in the amniotic membrane group were significantly higher than those in the rhEGF group and the control group (P0.05 or P0.01).Conclusion: human amniotic membrane can promote the healing of third degree burn of rabbit skin. In the early stage of wound healing, it can reduce the inflammatory reaction of wound by down-regulating the contents of IL-6, IL-8, IL-12 and TNF- 伪 in the wound tissue, upregulating the content of IL-10.
