镶嵌纳米丝素蛋白复合材料的制备及应用

【摘要】:院内感染是各医院内普遍存在的问题,主要是各种细菌感染,易造成患者的进一步健康损害。为此,开发具有抗菌性能的医疗器械表面涂装材料、设备具有重要意义。 从丝素蛋白作为保护蚕顺利度过蛹期而羽化成蛾的生理功能来看,丝素蛋白具有微调节周围环境的功用,,可抑制细菌繁殖,具备抑菌性。但是纯再生蚕丝素蛋白材料的成型性及柔韧性能较差,且价格昂贵,其应用受到限制。本课题拟将生物相容性良好的蚕丝素蛋白与成型性好的聚乙烯醇结合起来,开发一种具抗菌性的新型材料。研究采用高浓度的盐溶液溶解丝素纤维,并用中性蛋白酶酶解制备纳米级丝素蛋白(SFP),再采用水相共混,将所制得的丝素与聚乙烯醇(PVA)按照不同配比共混并成型,制备出了纳米丝素蛋白和聚乙烯醇的共混膜(SFP/PVA)。对SFP/PVA共混膜进行了力学性能的表征,证明了SFP/PVA共混膜的拉伸强度、能承受的最大力与断裂强度均较好。其中SFP/PVA配比为30/70的膜的力学性能最好。采用平板菌落计数的方法对其进行了抗菌性能测试,结果显示部分比例的SFP/PVA共混膜对大肠杆菌、金黄色葡萄球菌的抗菌率可达到60%以上,而对铜绿色假单胞菌的抗菌效果也可达到30%以上。血液相容性试验结果表明SFP/PVA共混膜的溶血率低于5%,符合生物材料溶血试验要求;并且SFP/PVA共混膜具备一定的抗凝血效果;血小板消耗率低于20%,符合与血液接触的材料的使用要求。SFP/PVA配比为30/70的膜的血液相容性比其它几组配比好。细胞毒性试验结果表明SFP/PVA共混膜均无细胞毒性,且SFP/PVA配比为30/70的膜具有促细胞生长的作用。初步证明了SFP/PVA共混膜作为医疗器械表面涂装材料或医用导管材料的应用前景。
[Abstract]:Nosocomial infection is a common problem in hospitals. Therefore, it is of great significance to develop surface coating materials for medical devices with antibacterial properties. As a physiological function to protect silkworm from pupal stage, silk fibroin has the function of microregulating the surrounding environment, inhibiting the propagation of bacteria and having bacteriostasis. However, the pure regenerated silk protein material has poor formability and flexibility, and its application is limited because of its high price. In this paper, a new antibacterial material was developed by combining silk fibroin with polyvinyl alcohol with good biocompatibility. In this paper, silk fibroin fibers were dissolved in high concentration salt solution and prepared by enzymatic hydrolysis of neutral protease. The prepared silk fibroin and polyvinyl alcohol (PVA) were blended and formed according to the different ratio of silk fibroin and polyvinyl alcohol (PVA), and then water phase was used to blend the silk fibroin and polyvinyl alcohol (PVA). A blend of nano-silk fibroin and polyvinyl alcohol was prepared. The mechanical properties of the SFP/PVA blend film were characterized. It was proved that the tensile strength of the SFP/PVA blend film was the most powerful and the breaking strength of the SFP/PVA blend film was better. The film with SFP/PVA ratio of 30 / 70 has the best mechanical properties. The antibacterial properties of SFP/PVA blend membrane were tested by the method of plate colony count. The results showed that the antibacterial rate of Staphylococcus aureus could reach more than 60% against Escherichia coli. The antimicrobial effect against Pseudomonas copper-green was more than 30%. The results of blood compatibility test showed that the hemolysis rate of SFP/PVA blend membrane was lower than 5, which met the requirements of biomaterial hemolysis test, and the SFP/PVA blend membrane had a certain anticoagulant effect. The platelet consumption rate was lower than 20%, which met the requirements of the use of materials in contact with blood. The membrane with a ratio of 30 / 70 to SFP / PVA had better blood compatibility than the other groups. The results of cytotoxicity test showed that the SFP/PVA blend membrane had no cytotoxicity, and the membrane with SFP/PVA ratio of 30 / 70 had the effect of promoting cell growth. The application prospect of SFP/PVA blend film as medical instrument surface coating material or medical conduit material was preliminarily proved.
