纯镁含锌微弧氧化涂层对成骨细胞生物活性的影响

【摘要】:目的:将纯镁超声微弧氧化后进行化学镀锌,对涂层进行成骨细胞生物相容性检测,评价纯镁微弧氧化经化学镀锌的生物涂层的生物相容性。方法:将纯镁制备成φ10mm×1.5mm的圆形镁片,超声微弧氧化后,进行化学镀锌,制备出表面含锌的微弧氧化纯镁涂层材料。实验分为三组:A组为超声微弧氧化组(MAO),B组为化学镀锌10min的MAO纯镁(MAO-Zn),C组为碱处理后化学镀锌10min的MAO纯镁(MAO-NaOH-Zn),(A组为对照组,B组及C组为实验组)用扫描电镜(SEM)和能谱分析(EDS)分别测量与研究各组试件表面的形态与成分,激光共聚焦显微镜观察成骨细胞的早期形态,用cck8法检测细胞增殖,并检测成骨细胞ALP活性。结果:扫描电镜观察经超声微弧氧化处理的试件表面粗糙,多孔,起伏不平,涂层的主要元素有Mg、O、Si、Zn。扫描电镜和激光共聚焦显示C组和B组表面细胞的生长和黏附均优于A组,C组优于B组。CCK-8和ALP的检测结果显示三组材料表面细胞的增殖、活性为C组B组A组,三组的比较差异均具有统计学意义。结论:含锌MAO涂层的镁具有良好的生物相容性,而经碱处理后的镀锌涂层生物相容性最佳。
[Abstract]:Aim: to evaluate the biocompatibility of electroless zinc plating coating on pure magnesium by ultrasonic microarc oxidation. Methods: pure magnesium was prepared into 蠁 10mm 脳 1.5mm circular magnesium sheet. After ultrasonic micro-arc oxidation, electroless zinc plating was carried out to prepare pure magnesium coating material containing zinc on the surface. The experiment was divided into three groups: group A: ultrasonic micro-arc oxidation group, group B, electroless galvanized 10min, MAO pure magnesium, MAO-ZnOZN, group C, MAO, pure magnesium, MAO, pure magnesium, 10min, after alkali treatment, control group, group B, and group C, as control group, group B and group C as experimental groups) and energy spectrum scores. The surface morphology and composition of each group of specimens were measured and studied by EDS. The early morphology of osteoblasts was observed by laser confocal microscopy. The proliferation and ALP activity of osteoblasts were detected by cck8 assay. Results: the surface of the samples treated by ultrasonic micro-arc oxidation was rough, porous and undulating, and the main elements of the coating were MgO / SiZn. The results of scanning electron microscopy and laser confocal scanning showed that the growth and adhesion of surface cells in group C and group B were superior to those in group A. CCK-8 and ALP showed that the proliferation of surface cells in group C was better than that in group B, and the activity of cells in group C was group A. The differences among the three groups were statistically significant. Conclusion: the magnesium coating containing zinc has good biocompatibility, but the zinc coating after alkali treatment has the best biocompatibility.
