钛酸钙涂层的生物相容性及动物实验研究

【摘要】:目的:本研究通过在体动物实验,研究钛酸钙“原位”涂层生物相容性,比较钛酸钙涂层、羟基磷灰石涂层及无涂层钛材料的骨结合能力,探索钛酸钙涂层钛材料作为新型体内植入物的可行性。 方法:选取48只新西兰大白兔,将钛酸钙涂层螺钉、羟基磷灰石螺钉及无涂层螺钉分别植入股骨内髁、外髁,分别于2、4、8、12周进行影像学检测、组织学检测和生物力学检测,观察所研究材料在动物体内与骨组织结合情况,并进行相关分析。 结果:(1)一般指标:新西兰兔饮食良好,2-3天后行动良好,无跛行,伤口愈合均为甲级,无明显并发症。(2)影像学检测:X线照片显示各时间点的螺钉与周围骨组织无放射线透亮区,植入的螺钉均留在原来的位置,无脱落,各股骨髁均没有出现骨折。(3)组织学检测:光学显微镜和扫描电镜检测,钛酸钙涂层和羟基磷灰石涂层螺钉周围骨组织排列均较致密,涂层与骨组织之间结合紧密,无纤维组织层,无涂层螺钉与骨组织之间存在纤维组织层。(4)生物力学测试:通过测得螺钉扭入、扭出时的扭力计算固定指数,比较后发现钛酸钙涂层与骨结合强度明显优于无涂层的钛表面,但弱于羟基磷灰石涂层。 结论:钛酸钙具有很好的组织相容性,与骨组织结合能力明显优于钛,与羟基磷灰石涂层生物相容性类似。钛酸钙涂层与钛基表面“原位”结合,不脱落,钛酸钙与骨组织可能存在化学键结合,是一种具有发展潜力的骨科种植体涂层材料。
[Abstract]:Objective: to study the biocompatibility of calcium titanate "in situ" coating and compare the bone binding ability of calcium titanate coating, hydroxyapatite coating and uncoated titanium material through in vivo animal experiments. To explore the feasibility of calcium titanate coated titanium as a new type of implant in vivo. Methods: calcium titanate coated screws, hydroxyapatite screws and uncoated screws were implanted into the medial and outer condyle of femur in 48 New Zealand white rabbits. The binding of the materials to bone tissue in vivo was observed and the correlation analysis was carried out. Results 1) General outcome measures: the New Zealand rabbits had good movement and no lameness after a good diet for 2-3 days. The wound healing was grade A, and there was no obvious complication. 2) the radiographic examination showed that the screws at each time point and the surrounding bone tissue had no radiography-transparent area. The implanted screws remained in their original position and did not fall off. No fracture was found in the condyle of the femur. Histological examination showed that the bone tissue around the screws was compact by optical microscope and scanning electron microscopy, and that around the calcium titanate and hydroxyapatite coated screws. The biomechanical test showed that there was a fibrous tissue layer between the coated screw and the bone tissue. The fixation index was calculated by measuring the torsion force of the screw in and out of the screw. It was found that the bonding strength of calcium titanate coating with bone was better than that of uncoated titanium surface, but weaker than that of hydroxyapatite coating. Conclusion: calcium titanate has good histocompatibility, and its binding ability to bone tissue is better than that of titanium, and the biocompatibility of calcium titanate is similar to that of hydroxyapatite coating. Calcium titanate coating is a kind of orthopedic implants coating material with potential development potential because it binds in situ with titanium substrate surface and does not fall off, and there may be chemical bond binding between calcium titanate coating and bone tissue.
