2种骨内植入材料界面的多核巨细胞分布与骨桥蛋白表达

【摘要】:目的:比较医用纯钛和聚甲基丙烯酸甲酯(PMMA)2种材料-骨界面多核巨细胞分布情况和骨桥蛋白表达情况。方法:将医用纯钛和PMMA制成的种植体各30枚植入30只SD大鼠的两侧股骨远端髁突内,分别于3、7、14、21和28d时处死动物。制备脱钙石蜡切片,HE染色观察种植体-骨间隙愈合界面多核巨细胞的分布;免疫组织化学法检测种植体-骨界面骨桥蛋白的表达情况。结果:HE结果显示在5个时间点里均可以观察到种植体-骨界面有多核巨细胞的形成,3d时达到高峰且PMMA组多核巨细胞的数量明显多于钛组(P0.05);多核巨细胞数目随时间推移均呈现下降趋势,在21d和28d时无统计学差异。骨桥蛋白的表达在7d时达到高峰期,且医用纯钛组明显强于PMMA组(P0.05)。随时间延长,骨桥蛋白在钛组和PMMA组的表达均逐渐减弱。结论:不同骨内植入材料影响其界面的多核巨细胞分布与骨桥蛋白的表达。
[Abstract]:Aim: to compare the distribution of multinucleated giant cells and osteopontin expression between pure titanium and polymethyl methacrylate (PMMA). Methods: 30 implants made of pure titanium and PMMA were implanted into the distal femoral condyle of 30 SD rats. The distribution of multinucleated giant cells in implant-bone gap healing interface was observed by HE staining and the expression of osteopontin in implant-bone interface was detected by immunohistochemical method. Results the size of multinucleated giant cells in the implant-bone interface reached a peak at 3 days after the implantation, and the number of multinucleated giant cells in the PMMA group was significantly higher than that in the titanium group (P0.05), and the number of multinucleated giant cells in the PMMA group was significantly higher than that in the titanium group (P0.05). All of them showed a downward trend. There was no statistical difference between 21 d and 28 d. The expression of osteopontin reached its peak at 7 days, and the medical pure titanium group was significantly stronger than PMMA group (P0.05). Over time, the expression of osteopontin in both titanium and PMMA groups decreased gradually. Conclusion: the distribution of multinucleated giant cells and the expression of osteopontin were affected by different intraosseous implants.
