不同方法制备的DDM异位成骨的实验研究

【摘要】:目的: 应用异位成骨实验检测不同方法制备的脱矿牙本质基质(Demineralized Dental Matrix,DDM)的骨诱导性及生物相容性,为下一阶段DDM骨诱导机理的深入研究做基础。 方法: 采用统一的取材标准收集各类离体牙,应用目前国外两种常规制备方法制备脱矿牙本质基质(DDM)。然后在4只Beagle犬双侧股部肌肉建立DDM肌内异位植入模型,实验根据植入材料的不同,分为A、B、 C、 D、 E五组,A组植入DDM1, B组植入DDM1’(灭活法处理的DDM1),C组植入DDM2, D组植入DDM2’(灭活法处理的DDM2),E组植入DDM3(成品DDM)。按预定位置在左侧植入A、B两组材料,右侧植入C、D、E三组材料,每组4个时间位点。术后分别于2W、4W、6W、8W取材,行大体、组织学观察及碱性磷酸酶活性测定,运用SPSS17.0统计软件进行统计学分析。 结果: 1、大体观察显示:所有Beagle犬各时间点术后饮食、活动均正常,无死亡,创口愈合良好,无红肿、脓性渗出及创口裂开,一周左右创口完全愈合。 2、各时间点的取材组织观察:所有实验组与对照组材料与肌肉之间结合较紧密,未见明显的纤维囊壁形成,肌肉组织正常,未见充血水肿,无感染、坏死。 3、组织学观察:DDM3植入比格犬股部肌袋内发生异位成骨:8周时有类骨质形成,其中可见小梁骨;DDM1、DDM2及DDM灭活组未见成骨发生。 4、ALP测定:组内比较,DDM3各时间点ALP的差异有统计学意义(P0.01),其余组各时间点ALP的差异无统计学意义。组间比较,DDM3组各时间点ALP值均高于其他组,差异有统计学意义(P0.01)。 结论: 在本实验条件下的观察周期内,自制的DDM及灭活的DDM具有良好的生物相容性,但未表现出骨诱导活性。DDM3(成品DDM)具有良好的生物相容性和骨诱导活性。
[Abstract]:Aim: to detect the bone induction and biocompatibility of demineralized dentin matrix (Demineralized Dental Matrix,DDM prepared by different methods by ectopic osteogenesis test, and to provide a basis for further study of bone induction mechanism of DDM in the next stage. Methods: all kinds of isolated teeth were collected by unified material collection standard, and demineralized dentin matrix (DDM). Was prepared by two conventional preparation methods abroad at present. Then the model of DDM intramuscular ectopic implantation was established in the bilateral thigh muscles of 4 Beagle dogs. According to the different implantation materials, the rats were divided into five groups: group A, group B, implantation of DDM1', group DDM1), C, group DDM2, D, group DDM2', group DDM2), E, group DDM3 (finished DDM). According to the predetermined position, A and B were implanted on the left, C, D and E were implanted on the right, with 4 time sites in each group. After operation, the samples were taken at 2W, 4W, 6W, 8W, gross, histologic observation and alkaline phosphatase activity were measured, and the statistical analysis was carried out by SPSS17.0 statistical software. Results: 1. General observation showed that all Beagle dogs had normal diet, no death, good wound healing, no redness and swelling, purulent exudation and wound laceration, and the wound healed completely in about one week. 2. Observation of the tissue at each time point: all the experimental group and the control group had close binding between the material and muscle, no obvious formation of fiber capsule wall, normal muscle tissue, no hyperemia and edema, no infection and necrosis. 3. Histologic observation: ectopic osteogenesis occurred in the thigh muscle bag of Beagle dogs after implantation of DDM3: osteoid formation was found at 8 weeks, but no osteogenesis was found in DDM1,DDM2 and DDM inactivated groups. 4, ALP determination: there was significant difference in ALP at each time point of DDM3 in the group (P 0.01), but there was no significant difference in ALP in the other groups at each time point. Compared with the other groups, the ALP value of DDM3 group was higher than that of other groups at each time point, and the difference was statistically significant (P 0.01). Conclusion: during the observation period, the self-made DDM and inactivated DDM have good biocompatibility, but do not show bone induction activity. DDM3 (finished DDM) has good biocompatibility and bone induction activity.
