耳蜗内毛细胞传入神经突触对氨基糖苷类抗生素毒性刺激的反应

【摘要】:目的 观察腹腔注射氨基糖苷类抗生素(硫酸庆大霉素)后C57BL/6J小鼠内毛细胞传入神经突触的形态和功能变化,并进一步研究这种变化和听力改变的关系。 方法 选择出生后5周大小的C57BL/6J小鼠,雌雄不限,每组由6只小鼠组成,每天每只小鼠腹腔注射硫酸庆大霉素,药物浓度为100mg/kg,连续给药14d,然后停药后饲养至第28d.分别选取第4d、第7d、第10d、第14d和第28d的小鼠进行ABR(ClickTone burst)和DPOAE检测,未进行腹腔注射给药的小鼠(0d)作为对照组,对照组动物每天给予相同剂量的生理盐水。动物处死后耳蜗取材,然后分别对实验组和对照组小鼠进行基底膜铺片染色和蛋白检测,以观察耳蜗内毛细胞传入神经突触前特异性蛋白RIBEYE/CtBP2和突触后神经递质GluR2/3受体的表达情况。 结果 应用庆大霉素以后,,实验组小鼠从第4d开始出现听力损失,在第7d时听力损失似乎达到最明显的水平,而第10d,14d,28d听力阈值较第7d时有所降低,但并无显著性差异(P>0.05),而实验组小鼠在第10d,14d,28d的听力阈值仍显著高于对照组(P<0.05),在应用庆大霉素后的第4d,7d,10d,14d和28d, RIBEYE/CtBP2和GluR2/3在基底膜上的表达水平和部位均出现了明显的变化,而且这种变化和小鼠听阈的改变存在一定的对应关系。 结论 一定剂量的庆大霉素所造成的听力损害可能和内毛细胞传入神经突触的形态及功能改变有关,而且该突触形态上的恢复可能有助于小鼠听力的改善。
[Abstract]:Purpose. To observe the morphological and functional changes of afferent synapses in C57BL / 6J mice after intraperitoneal injection of aminoglycoside antibiotics (gentamicin sulfate), and to further study the relationship between this change and hearing changes. Method. Five weeks after birth, C57BL / 6J mice, male and female, were randomly divided into six groups. Each mouse was intraperitoneally injected with gentamicin sulfate every day. The drug concentration was 100 mg / kg for 14 days, and then was fed to 28 days after withdrawal. The mice of 4 days, 7 days, 10 days, 14 days and 28 days were tested for ABR(ClickTone burst and DPOAE, and the mice that were not injected intraperitoneally for 0 d) were used as control group. The rats in the control group were given the same dose of normal saline every day. The cochlea were collected after the animals were killed, and then the mice in the experimental group and the control group were stained with basement membrane and tested for protein. To observe the expression of presynaptic specific protein (RIBEYE/CtBP2) and postsynaptic neurotransmitter (GluR2/3) receptor of afferent nerve in cochlear hair cells. Results. After application of gentamicin, the hearing loss in the experimental group appeared from the 4th day, and appeared to reach the most obvious level on the 7th day, and the threshold of hearing loss on the 10th day after 14 days was lower than that on the seventh day. However, there was no significant difference (P > 0.05), but the hearing threshold of the experimental group was significantly higher than that of the control group on the 10th day, 14d and 28d, and the expression of RIBEYE/CtBP2 and GluR2/3 on the basement membrane showed obvious changes on the 4th day, 7th day, 10d, 14d and 28d after gentamicin administration, but the hearing threshold of the experimental group was significantly higher than that of the control group (P < 0.05). Moreover, there is a certain correspondence between the change of hearing threshold and the change of hearing threshold in mice. Conclusion. The hearing loss caused by gentamicin at a certain dose may be related to the morphological and functional changes of the afferent synapse of the inner hair cell, and the morphological recovery of the synapse may contribute to the improvement of the hearing ability of the mice.
