注射神经生长因子对兔眼外肌的影响

【摘要】:目的观察大白兔眼外肌注射神经生长因子(NGF)对眼外肌的影响。方法将8只大白兔右眼设为观察组,左眼设为对照组,观察组眼上直肌注射10μg NGF;对照组眼上直肌相同位置注射等量生理盐水。测量眼外肌湿重,计算眼外肌横截面积,HE染色测量肌纤维周径、观察炎症情况,免疫组化染色测定活化卫星细胞。结果观察组上直肌横截面积[(12.412±1.372)mm2]显著大于对照组[(6.738±0.856)mm~2,P0.05]。观察组兔肌纤维周径[(228.32±16.57)μm]显著大于对照组[(129.32±11.16)μm,P0.05]。观察组大、中、小细胞构成比分别为27.0%、69.6%、3.5%,对照组大、中、小细胞构成比分别为13.4%、63.5%、23.2%,两组比较差异有统计学意义(P0.05)。HE染色:治疗后2 w,两组兔眼外肌均没有炎症细胞浸润和细胞坏死,肌纤维边界清楚、方向一致、排列整齐。免疫组化染色:观察组活化卫星细胞百分率[(28.78±4.36)%]显著大于对照组[(11.87±2.47)%,P0.05]。结论眼外肌注射NGF能够活化卫星细胞,促进眼外肌肌纤维增生,对眼组织没有明显不良影响。
[Abstract]:Objective to observe the effect of nerve growth factor (NGF) injection into the extraocular muscles of rabbits. In the observation group, the superior rectus muscle was injected with 10 渭 g NGF and the control group was injected with the same amount of normal saline at the same position. The wet weight of the extraocular muscle was measured, the cross sectional area of the extraocular muscle was calculated and the diameter of the muscle fiber was measured by HE staining, and the inflammation was observed. Results the cross sectional area of the superior rectus muscle in the observation group [12.412 卤1.372)mm2] was significantly larger than that in the control group [6.738 卤0.856 mm]. The peripheral diameter of muscle fiber in the observation group [228.32 卤16.57 渭 m] was significantly higher than that in the control group [129.32 卤11.16 渭 m P0.05]. The ratio of large, middle and small cells in the observation group was 27.00.69.63.5 渭 m, and that in the control group was larger than that in the control group. The ratio of small cells to small cells was 13. 4% and 63. 5% and 23. 2, respectively. The difference between the two groups was statistically significant (P 0. 05). He staining: at 2 weeks after treatment, there were no inflammatory cell infiltration and cell necrosis in the extraocular muscles of the two groups, and the muscle fiber boundary was clear and the direction was the same. Immunohistochemical staining: the percentage of activated satellite cells in the observation group [28.78 卤4.36] was significantly higher than that in the control group [11.87 卤2.47]. Conclusion injection of NGF into the extraocular muscle can activate the satellite cells, promote the proliferation of the muscle fiber of the extraocular muscle, and have no obvious adverse effect on the ocular tissue.
