MicroRNA在急性前葡萄膜炎中的遗传易感性研究

【摘要】:目的本研究旨在探讨micro RNA(mi RNA)拷贝数变异与急性前葡萄膜炎伴或不伴发强直性脊柱炎之间的遗传相关性以及其潜在的发病机制。方法本实验共纳入768例急性前葡萄膜炎伴或不伴发强直性脊柱炎的患者及660例正常人对照。我们采用Taq Man PCR法检测mi RNA基因拷贝数分型,实时荧光定量PCR(Real-Time PCR)检测mi RNA的m RNA表达水平,流式细胞术和ELISA法分别用来检测mi R~(-9)-3的转染效率以及细胞因子的表达水平。结果低拷贝(CNV2)的mi R-143、mi R-146a、mi R~(-9)-3、mi R-205以及高拷贝(CNV2)的mi R-301a和mi R-23a频率在伴发强直性脊柱炎的急性前葡萄膜炎患者中明显增高(P=3.725×10~(-5)至8.033×10~(-9))。我们还发现,mi R-146a低拷贝频率和mi R-23a、mi R-205的高拷贝频率与不伴发强直性脊柱炎的急性前葡萄膜炎呈显著性相关(P=0.002至P=0.001)。与正常人相比,mi R~(-9)-3,mi R-143和mi R-146a的m RNA表达水平在伴发强直性脊柱炎的急性前葡萄膜炎患者中显著增高。另外,ELISA检测发现IL-1β和IL-6在人视网膜色素上皮(h RPE)细胞中的表达量可能是由mi R~(-9)-3调控的。结论低拷贝的mi R-143、mi R-146a、mi R~(-9)-3和mi R-205以及高拷贝的mi R-301a和mi R-23a与伴发强直性脊柱炎的急性前葡萄膜炎遗传易感性相关。低拷贝的mi R-146a和高拷贝的mi R-23a和mi R-205与不伴发强直性脊柱炎的急性前葡萄膜炎的发病呈相关性。
[Abstract]:Objective to investigate the genetic relationship between micro RNA(mi copy number variation and acute anterior uveitis with or without ankylosing spondylitis and its underlying pathogenesis. Methods 768 cases of acute anterior uveitis with or without ankylosing spondylitis and 660 normal controls were included. The Taq Man PCR method was used to detect the copy number typing of mi RNA gene, the m RNA expression level of mi RNA was detected by real-time fluorescence quantitative PCR(Real-Time PCR, and the transfection efficiency and cytokine expression level of mi RNA were detected by flow cytometry and ELISA, respectively. Results the frequency of mi R-301a and mi R-23a in low-copy CNV2) patients with acute anterior uveitis with ankylosing spondylitis was significantly higher than that in patients with acute anterior uveitis (P 3.725 脳 10 ~ (-5) to 8.033 脳 10 ~ (10). We also found that the low copy frequency of mi-mi R-146a and the high copy frequency of mi R-23aanmi R-205 were significantly correlated with acute anterior uveitis without ankylosing spondylitis. Compared with the normal controls, the expression levels of m RNA in the mitomyr-9ke-3a R-143 and mi R-146a were significantly higher in the patients with acute anterior uveitis associated with ankylosing spondylitis (ankylosing spondylitis). In addition, the expression of IL-1 尾 and IL-6 in human retinal pigment epithelium (RPE) cells was detected by Elisa. Conclusion the low copy of mi R-143Li mi R-146a mi r-146a mi r-9- 3 and mi R-205 and the high-copy miR-301a and miR-23a are associated with the genetic susceptibility of acute anterior uveitis with ankylosing spondylitis (ankylosing spondylitis). Low copy mi R-146a and high copy mi R-23a and mi R-205 were associated with acute anterior uveitis without ankylosing spondylitis.
