5-氨基酮戊酸光动力学对幼年型喉乳头状瘤移植瘤的杀伤作用的研究

【摘要】:目的:研究5-氨基酮戊酸光动力学(ALA-PDT)对裸鼠幼年型喉乳头状瘤移植瘤的杀伤作用。 方法:建立裸鼠幼年型喉乳头状瘤移植瘤模型,在肿瘤局部应用5~氨基酮戊酸溶液,3小时后用波长为630nm的氮氖激光照射肿瘤10min,照射剂量为90J/Cm2。辐照后,每天观察肿瘤变化情况,记录测量肿瘤体积并计算相对肿瘤增殖率T/C,并设单用5-ALA,单用PDT,阴性对照组,观察处理后的肿瘤的组织形态学变化。采用原位杂交、TUNEL检测、透射电镜的方法,研究5-氨基酮戊酸光动力对喉乳头状瘤组织细胞凋亡情况的影响,应用荧光定量聚合酶链反应(polymerase chain reaction,PCR)法检测肿瘤组织中的HPV6/11含量。 结果:成功建立幼年型喉乳头状瘤手术标本组织块移植瘤模型。5-ALA-PDT治疗组的肿瘤组织均可使肿瘤显著缩小,肿瘤表面可见坏死,TUNEL检测及透射电镜均见肿瘤组织有广泛凋亡及坏死,24h及48h的相对肿瘤增值率分别是11.6%和17.2%,p0.05,荧光定量PCR检测病毒含量明显减少。生理盐水对照组、5-ALA组、PDT组对肿瘤无显著作用。 结论:5-ALA-PDT对幼年型喉乳头状瘤的生长有显著的抑制作用。
[Abstract]:Aim: to study the killing effect of 5 aminolevulinic acid (ALA-PDT) on juvenile laryngeal papilloma in nude mice. Methods: the transplanted tumor model of juvenile laryngeal papilloma in nude mice was established. After 3 hours of local application of 5-aminolevulinic acid solution, the tumor was irradiated with 630nm laser for 10 min at a dose of 90 J / cm ~ 2. After irradiation, tumor changes were observed daily, tumor volume was measured and relative tumor proliferation rate (T / C) was calculated. The histomorphologic changes of the treated tumors were observed by 5-ALA-PDT alone and PDT-negative control group. The effect of 5-aminolevulinic acid on apoptosis of human laryngeal papilloma tissue was studied by in situ hybridization Tunel and transmission electron microscopy. The HPV6 / 11 content in tumor tissue was detected by fluorescence quantitative polymerase chain reaction (FQ-PCR) and (polymerase chain reaction-polymerase chain reaction (FQ-PCR). Results: the tumor tissue of the group treated with 5-ALA-PDT could significantly reduce the tumor size, and the model of transplanted tumor was established successfully in the operation specimen of juvenile laryngeal papilloma. TUNEL-mediated dUTP Nick end labeling (Tunel) and transmission electron microscopy (TEM) showed that the relative tumor proliferation rates of 24 h and 48 h were 11.6% and 17.2%, respectively, and the amount of virus detected by fluorescence quantitative PCR was significantly decreased. Normal saline control group, 5-ALA group, PDT group had no significant effect on tumor. Conclusion the growth of juvenile laryngeal papilloma was significantly inhibited by 5-ALA-PDT.
