ERK2及p-ERK2在鼻息肉组织的表达

【摘要】:目的:1、检测细胞外信号调节激酶2(ERK2)及磷酸化细胞外信号调节激酶2(p-ERK2)在鼻息肉(Nasal polyp,NP)组织的表达情况;2、初步探讨ERK2/p-ERK2在NP发病中的临床意义。方法:组织标本收集于山西医科大学第二医院耳鼻咽喉头颈外科行鼻内镜手术的慢性鼻窦炎伴鼻息肉患者(Chronic rhinosinusitis with nasal polyps,CRSwNP)(根据EPOS2012,鼻窦炎诊治更新与精要)30例,起止时间为2016年6月至2016年12月期间。术中取CRSwNP患者NP标本30份(实验组),同时取该患者下鼻甲正常鼻黏膜组织标本30份(对照组)。应用间接免疫荧光技术(Indirect immunofluorescence,IIF)检测ERK2/p-ERK2在NP及正常鼻黏膜组织的表达,并探讨其临床意义。结果:1、NP:p-ERK2主要表达在增生的上皮细胞、炎症细胞以及腺上皮细胞,主要位于胞核;ERK2主要表达在腺上皮细胞、炎症细胞,主要位于胞质。正常鼻黏膜组织:p-ERK2主要表达在炎症细胞、腺上皮细胞,主要位于胞核;ERK2主要位于炎症细胞,主要位于胞质。2、p-ERK2在NP和正常鼻黏膜组织的表达有统计学差异(P0.05);ERK2在NP和正常鼻黏膜组织的表达无统计学差异(P0.05)。结论:1、p-ERK2在NP的发病中可能具有重要作用,而ERK2与NP的发生可能无关。2、ERK2在NP和正常鼻黏膜组织均有表达,提示ERK2可能参与鼻黏膜正常生理功能的维持。3、RAF-1-MEK-ERK2-NF-KB信号通路可能参与NP的发病。
[Abstract]:Objective to investigate the expression of extracellular signal-regulated kinase 2 (ERK2) and phosphorylated extracellular signal-regulated kinase 2 (p-ERK2) in nasal polyp (NP), and to explore the clinical significance of ERK2 / p-ERK2 in the pathogenesis of NP. Methods: tissue specimens were collected from 30 patients with chronic sinusitis associated with nasal polyps (according to EPOS2012) who underwent endoscopic sinus surgery in the Department of Otorhinolaryngology and head and neck surgery, Shanxi Medical University, and 30 patients with chronic sinusitis and nasal polyps (according to EPOS2012, update and essentials of the diagnosis and treatment of sinusitis), 30 patients with chronic sinusitis and nasal polyps were studied. The starting and ending period is from June 2016 to December 2016. Thirty NP specimens of CRSwNP patients (experimental group) and 30 specimens of normal nasal mucosa of inferior turbinate (control group) were taken during operation. The expression of ERK2 / p-ERK2 in NP and normal nasal mucosa was detected by indirect immunofluorescence technique (IIF) and its clinical significance was discussed. Results the expression of p-ERK2 was mainly in proliferative epithelial cells, inflammatory cells and glandular epithelial cells, mainly in glandular epithelial cells, inflammatory cells and cytoplasm. The normal nasal mucosa: p-ERK2 is mainly expressed in inflammatory cells, glandular epithelial cells, mainly located in nuclear ERK2, mainly in inflammatory cells. The expression of ERK2 in NP and normal nasal mucosa was significantly different (P0.05), and there was no significant difference in the expression of ERK2 between NP and normal nasal mucosa (P0.05). Conclusion 1 / 1 p-ERK2 may play an important role in the pathogenesis of NP, but ERK2 may not be related to the occurrence of NP, and ERK2 may be expressed in both NP and normal nasal mucosa. These results suggest that ERK2 may be involved in the maintenance of normal physiological function of nasal mucosa. 3RAF-1-MEK-ERK2-NF-KB signaling pathway may be involved in the pathogenesis of NP.
