羊膜泪道支架预防去势雄兔干眼症的实验研究

【摘要】:目的探究羊膜泪道支架对雄兔干眼症的预防效果。方法选取36只健康雄兔,剪除第三眼睑并抗感染处理,随机分为3组(每组12只),制作去势雄兔模型,其中A组为阴性对照组,B组为干眼模型组,C组为泪道羊膜预防组,并在植入羊膜泪道支架前、植入后2周、4周及6周行角膜荧光素(fluorescent,FL)染色、Schirmer I试验、Western blot、免疫荧光染色及角膜共聚焦显微镜检查。结果三组不同时间点间的泪液分泌量及FL有明显改变(F=7.126,P=0.009;F=9.658,P=0.016),三组间泪液分泌量及FL有明显改变(F=12.582,P=0.005;F=13.187,P=0.013),三组的泪液分泌量及FL变化趋势也有明显改变(F=8.531,P=0.007;F=10.652,P=0.019)。A、B、C三组植入后6周上皮基底细胞分别(3811±414)个细胞、(3820±314)个细胞、(2789±353)个细胞,炎症细胞密度分别为(266±28)个细胞、(266±29)个细胞、(67±13)个细胞;A、B、C三组植入后6周上皮基底细胞(F=13.442,P=0.012)和炎症细胞密度(F=9.231,P=0.021)比较,差异均有统计学意义。泪道上皮K16染色均为阴性,植入羊膜泪道支架后各时间点A、B、C三组兔泪小管组织中α-SMA表达无明显改变,差异有统计学意义(F=14.681,P=0.002)。结论植入羊膜泪道支架对雄兔干眼症的预防有一定的效果。
[Abstract]:Objective to investigate the preventive effect of amniotic lacrimal duct stent on dry eye in male rabbits. Methods Thirty-six healthy male rabbits were randomly divided into 3 groups (12 in each group), with the third eyelid being cut off and treated with anti-infection. The ovariectomized male rabbit model was established in which group A was negative control group and group B was dry eye model group, group C was lacrimal duct amniotic membrane prophylaxis group. Before and after implantation of amniotic lacrimal duct stent, fluorescentin FL staining and Western blot, immunofluorescence staining and corneal confocal microscopy were performed 2 weeks and 6 weeks after implantation. Results there were significant changes in tear secretion and FL in the three groups at different time points (FF7.126P0.009 / F9.658 / P0. 016). There were significant changes in tear secretion and FL between the three groups (FF12. 582PU 0. 005 FN 13.187 P0.013). There was also a significant change in tear secretion and FL in the three groups (FF8. 531P0. 007FN 10. 652P0.019). ABC group 3 had a significant change in the epithelium of the epithelium 6 weeks after implantation (FF8. 531P0. 007FN 10.652P0. 019). Basal cells were (3811 卤414) cells, (3820 卤314) cells and (2789 卤353) cells, respectively. The density of inflammatory cells was (266 卤28) cells, (266 卤29) cells, and (67 卤13) cells, respectively. There were significant differences in epithelial basal cells (F _ (13.442) P _ (0.012) and inflammatory cell densities (F _ (9.231) P _ (0.021) 6 weeks after implantation. The expression of 伪 -SMA in lacrimal canaliculus tissues of the three groups was not significantly changed after the implantation of amniotic lacrimal duct stents at all time points, and the difference was statistically significant (F _ (14.681) P ~ (0.002). Conclusion the implantation of amniotic lacrimal duct stent has certain effect on the prevention of xerophthalmia in male rabbits.
