microRNA-126在多能干细胞定向分化为视网膜神经干细胞过程中表达量的变化及意义

【摘要】:目的研究microRNA-126(miR-126)在湿性老年黄斑变性(wet AMD)疾病患者来源的多能干细胞(i PSC)定向分化为视网膜神经干细胞(RNSC)中表达量的变化及意义。方法选取新发现的wet AMD患者10例作为观察组,无眼底病变及全身病的10例作为对照组。抽取受试者的外周静脉血,提取单个核细胞(PBMC),进一步提取CD34+细胞,行腺病毒转染,诱导产生i PSC且进行鉴定;应用实时定量PCR(qRT-PCR)技术检测miR-126在10个不同诱导发育阶段的RNSC中的相对表达量。结果 wet AMD疾病患者的外周血成功诱导分化出RNSC;观察组miR-126的不同诱导时间相对表达量的差异除了时刻8、9、10其余测试时间点均有统计学意义(P0.05);对照组miR-126的不同诱导时间相对表达量的差异无统计学意义(P0.05);观察组与对照组细胞miR-126相对表达量在不同诱导时间的差异有统计学意义(P0.05)。结论 wet AMD患者早期miR-126的表达量显著升高,且有逐渐降低的趋势。
[Abstract]:Objective to study the changes and significance of microRNA-126 (miR-126) expression in the differentiation of multipotent stem cells (i PSC) into retinal neural stem cells (RNSC) in elderly patients with (wet AMD) disease of macular degeneration. Methods Ten newly discovered wet AMD patients were selected as observation group, 10 patients without fundus disease and 10 patients with systemic disease as control group. The peripheral venous blood was extracted and the (PBMC), of mononuclear cells was extracted. The CD34 cells were further extracted by adenovirus transfection and I PSC was induced and identified. Real time quantitative PCR (qRT-PCR) was used to detect the relative expression of miR-126 in 10 RNSC at different stages of induction and development. Results RNSCs were successfully induced and differentiated from peripheral blood of patients with wet AMD disease, and the difference of relative expression of miR-126 at different induction time in observation group was statistically significant (P0.05), and that in control group was different in different miR-126 induction time (P0.05). There was no significant difference in the relative expression of miR-126 between the two groups (P0.05); there was significant difference in the relative expression of miR-126 between the observation group and the control group at different induction time (P0.05). Conclusion the expression of miR-126 in the early stage of wet AMD patients increased significantly and tended to decrease gradually.
