息肉状脉络膜血管病变血清药物靶向因子的初步筛选

【摘要】:目的利用蛋白芯片技术筛选息肉状脉络膜(PCV)血清差异性蛋白,以便寻找PCV的早期诊断标志物和药物治疗新靶点,为进一步探讨PCV发病机制提供依据。方法1.用AAH-BLG-507蛋白芯片(含507种功能蛋白)检测随机抽取的1例阴性对照组、1例AMD患者及2例PCV患者血清中相应蛋白的表达水平,分析差异性表达蛋白。2.根据AAH-BLG-507蛋白芯片结果,选取相关细胞因子和趋化因子构建成含34种因子的多因子检测试剂盒(即液相芯片),检测65例PCV患者(男性42例,女性23例;年龄分布在49岁到81岁之间)和70例阴性对照组(男性40例,女性30例;年龄分布在53岁到78岁之间)血清中相关34种因子的表达,筛选有差异性表达的因子。结果1.入组的PCV患者平均年龄(65.43±8.57)岁,与对照组相比无显著差异(P=0.070.05)。两组性别构成比较,结果显示无显著差异(P0.05),认为两组患者年龄和性别匹配。2.AAH-BLG-507蛋白芯片检测结果分析显示,有79种蛋白表达有差异,其中58种在PCV患者血清中呈上调趋势,21种呈下调趋势。分析表达差异性蛋白之间关系,结果显示主要分为细胞因子、趋化因子、胞外基质蛋白/基质金属蛋白酶、生长因子及新生血管相关因子及其受体4大类。3.多因子检测试剂盒结果显示,选取的34种因子中有10种有统计学差异(P0.05)其中8种因子及其在PCV血清中对应的浓度:Eotaxin(52.618±24.954)pg/mL,IL-1RA(289.232±146.678)pg/m L;IL-1β(0.481±0.279)pg/mL、IL-4(2.069±1.050)pg/mL、IL-5(0.169±0.111)pg/mL、IL-13(0.424±0.122)pg/mL、IL-22(4.730±3.790)pg/m L、GM-CSF(1.542±0.752)pg/mL,明显高于其在对照组血清浓度,即这8种因子在PCV患者血清中呈上调趋势(P值分别为0.003、0.007、0.009、0.002、0.040、0.003、0.003、0.003均0.05)。而另外2种因子在PCV患者血清中浓度IL-7(2.929±2.749)pg/mL、SDF-1α(347.141±101.243)pg/m L明显低于对照组,即呈下调趋势(P值分别为0.003和0.008均0.05)。结论1.蛋白芯片技术是一种快速、简便的高通量蛋白质组研究方法,为PCV研究提供了新的研究策略。2.PCV的发病可能与患者血清中Eotaxin、IL-1RA、GM-CSF、IL-1β、IL-4、IL-5、IL-13、IL-22表达上调和IL-7、SDF-1α表达下调相关。3.筛选出的PCV相关的差异蛋白,将可能作为PCV患者的诊断分子标志物和药物治疗靶向因子。

Method 1. The expression levels of the corresponding proteins in the serum of 1 AMD patient and 2 PCV patients were detected by AAH-BLG-507 protein chip (including 507 functional proteins), and the differentially expressed protein. 2 was analyzed. The expression of differentially expressed proteins was analyzed.
