离子导入巩膜交联术对近视模型力学性能及胶原代谢的影响

【摘要】:巩膜胶原交联术是有望控制近视病程发展的方法之一。然而,该术式的确切程序及其对近视动物模型的影响仍有待探讨。将离子导入法引入巩膜核黄素-紫外线A(UVA)交联术,采用10mW/cm~2,9min的快速交联方案,研究该术式对于近视动物模型巩膜生物力学性能及胶原代谢的影响。利用形觉剥夺法将96只新西兰大白兔诱导形成相对稳定的近视模型,然后进行巩膜交联术或假手术,术后4个时间点(1d,10d,1个月,3个月)分别采集样本和获取数据。术后4个时间点,与非交联近视眼相比,交联近视眼巩膜的极限应力、杨氏模量和生理杨氏模量均明显增加。此外,交联组与未交联组胶原代谢相关基因和蛋白的表达存在显著的差异。结果表明,离子导入核黄素-紫外线A快速交联术很可能是一种控制近视病理过程的可行性方法。
[Abstract]:Scleral collagen crosslinking is one of the promising methods to control the progression of myopia. However, the exact procedure of this procedure and its effect on the animal model of myopia need to be discussed. The scleral riboflavin ultraviolet A (UVA) crosslinking was introduced into the scleral sclera. The effects of this procedure on the scleral biomechanical properties and collagen metabolism in myopic animal models were studied by a rapid crosslinking protocol of 10 MW / cm ~ 2 for 9 min. Ninety-six New Zealand white rabbits were induced to form a relatively stable myopia model by the method of form deprivation. Scleral crosslinking or sham-operation was performed. Samples were collected and data were obtained at 4 time points (1 day, 10 days, 1 month, 3 months) after operation. The scleral ultimate stress, Young's modulus and physiological Young's modulus of cross-linked myopia were significantly increased compared with non-cross-linked myopia at 4 time points after operation. In addition, there were significant differences in the expression of collagen metabolism-related genes and proteins between crosslinked and uncrosslinked groups. The results showed that the rapid cross-linking of riboflavin and ultraviolet A might be a feasible method to control the pathological process of myopia.
