缓激肽对体外培养大鼠血视网膜屏障通透性的影响

【摘要】:目的探讨缓激肽对体外培养大鼠血视网膜屏障通透性的影响。方法将缓激肽作用于Transwell小室建立体外培养的大鼠视网膜血管内皮细胞(REVC)间紧密连接屏障模型,通过跨上皮细胞电阻(TER)值观察缓激肽对大鼠REVC间紧密连接结构的影响;RT-PCR法检测紧密连接相关蛋白ZO-1和occludin mRNA表达水平的变化。结果实验组作用2 h的TER值小于测量前、作用4 h组和对照组(P0.01);实验组作用4 h组和测量前、对照组TER值无明显变化(P0.05)。结论缓激肽可在转录水平上减少紧密连接相关蛋白ZO-1和occludin mRNA表达,能够开放大鼠REVC间的紧密连接,增加血视网膜屏障的通透性。
[Abstract]:Objective to investigate the effect of bradykinin on the permeability of blood-retinal barrier in vitro. Methods bradykinin was applied to Transwell chamber to establish a rat model of tight junction barrier between (REVC) and vascular endothelial cells (RECs) cultured in vitro. The effect of bradykinin on the tight junction structure of rat REVC was observed by (TER) value of transepithelial cells. The expression levels of ZO-1 and occludin mRNA were detected by RT-PCR. Results the TER value of the experimental group at 2 h was lower than that of the control group (P0.01), and the TER value of the experimental group was not changed significantly (P0.05). Conclusion bradykinin can reduce the expression of ZO-1 and occludin mRNA at the transcriptional level, open the tight connection between REVC and increase the permeability of blood-retinal barrier.
