CD36在慢性鼻-鼻窦炎伴鼻息肉组织中的表达与意义

【摘要】:目的炎症在慢性鼻-鼻窦炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)的发生发展过程中起着重要作用,并且炎症在CRSwNP的病理机制仍然不完全清楚。此研究主要观察清道夫受体CD36在慢性鼻-鼻窦炎伴鼻息肉的表达。方法使用逆转录酶聚合酶链反应(RT-PCR),Western blot免疫印迹方法检测来自合计43例慢性鼻-鼻窦炎(chronic rhinosinusitis,CRS)或伴息肉患者的手术标本CD36的mRNA和蛋白表达。进一步通过免疫组织化学和免疫荧光技术观察CD36表达与分布规律。结果我们的RT-PCR和Western blot免疫印迹分析的结果表明,与正常鼻黏膜组织相比,鼻息肉标本中CD36 mRNA和蛋白表达显著增强(P0.01)。免疫组织化学结果显示,CD36弱表达于慢性鼻窦炎鼻黏膜组织,而鼻息肉组织中表达较强。CD36与血管内皮标记物CD31的双染荧光结果显示,CD36阳性的细胞与CD31的阳性共标。结论研究数据表明CD36在慢性鼻-鼻窦炎伴鼻息肉发生与发展过程可能起着重要作用。
[Abstract]:Objective inflammation plays an important role in the development of chronic rhinosinusitis with nasal polyp (chronic rhinosinusitis with nasal polyps,CRSwNP), and the pathological mechanism of inflammation in CRSwNP is still unclear. The purpose of this study was to investigate the expression of scavenger receptor CD36 in chronic rhinosinusitis with nasal polyps. Methods the expression of mRNA and protein in CD36 from 43 patients with chronic rhinosinusitis (chronic rhinosinusitis,CRS) or polyps was detected by reverse transcriptase polymerase chain reaction (RT-PCR) Western blot Western blot. The expression and distribution of CD36 were observed by immunohistochemistry and immunofluorescence technique. Results the results of RT-PCR and Western blot Western blot analysis showed that the expression of CD36 mRNA and protein in nasal polyps was significantly higher than that in normal nasal mucosa (P0.01). The results of immunohistochemistry showed that the expression of CD36 was weak in the nasal mucosa of chronic sinusitis, while the expression of CD36 in nasal polyps was stronger than that in the vascular endothelial marker CD31. The results showed that the cells with positive expression of CD36 were colabeled with CD31. Conclusion CD36 may play an important role in the occurrence and development of chronic rhinosinusitis with nasal polyps.
