Tau在鼻咽癌亲本及紫杉醇耐药细胞系中的表达差异

【摘要】:目的: 分析微管相关蛋白Tau在鼻咽癌亲本细胞及紫杉醇耐药细胞系中的表达差异,探讨Tau在鼻咽癌紫杉醇耐药中的作用。 方法: 以鼻咽癌亲本细胞株CNE-1、CNE-2及其相对应的紫杉醇耐药细胞株CNE-1/taxol、CNE-2/taxol为实验对象,分别应用Western Bolt及实时荧光定量PCR技术检测Tau在这些细胞中的蛋白和mRNA水平的表达。 结果: 以Western Bolt和实时荧光定量PCR的结果反映Tau在不同细胞系中蛋白和mRNA水平的表达。Tau蛋白的表达水平以相对光密度参考值,即Tau蛋白的光密度参考值/同一样本GAPDH的光密度参考值表示。mRNA的相对表达量用2-△Ct方法计算得出。结果得出,Tau蛋白在鼻咽癌亲本及紫杉醇耐药细胞系中均有表达,其中耐药细胞CNE-1/taxol、CNE-2/taxol分别是亲本细胞CNE-1、CNE-2的3.11±0.54,1.39±0.21倍。Tau的mRNA在各个鼻咽癌细胞中也有表达,耐药细胞CNE-1/taxol、CNE-2/taxol分别是亲本细胞CNE-1、CNE-2的2.48±0.33、1.67±0.15倍。 结论: Tau在鼻咽癌紫杉醇耐药细胞系中的表达高于亲本细胞系,Tau可能与紫杉醇耐药相关。Tau可以作为紫杉醇化疗敏感性及预后的个预测分子,对其进行干预可以在一定程度上逆转紫杉醇耐药。
[Abstract]:Aim: to investigate the expression of microtubule-associated protein (Tau) in nasopharyngeal carcinoma (NPC) parent cells and paclitaxel resistant cell lines, and to explore the role of Tau in taxol resistance of nasopharyngeal carcinoma (NPC). Methods: the expression of Tau protein and mRNA in nasopharyngeal carcinoma (NPC) parent cell line CNE-1,CNE-2 and its taxol resistant cell line CNE-1/taxol,CNE-2/taxol were detected by Western Bolt and real-time fluorescence quantitative PCR. Results: the results of Western Bolt and real-time quantitative PCR showed that the expression level of Tau protein and mRNA in different cell lines was the reference value of relative optical density. The optical density reference value of Tau protein and the optical density reference value of GAPDH in the same sample were calculated by 2-Ct method. The results showed that CNE-1/taxol,CNE-2/taxol protein was expressed in nasopharyngeal carcinoma (NPC) parents and paclitaxel resistant cell lines. The expression of mRNA in resistant cells was 3.11 卤0.54n1.39 卤0.21 times of parental CNE-1,CNE-2, respectively. Tau was also expressed in various nasopharyngeal carcinoma cells. The CNE-1/taxol,CNE-2/taxol of drug resistant cells was 2.48 卤0.33 1.67 卤0.15 times of that of parental CNE-1,CNE-2, respectively. Conclusion: the expression of Tau in paclitaxel-resistant nasopharyngeal carcinoma cell line is higher than that in parent cell line. Tau may be a predictor of paclitaxel chemosensitivity and prognosis. Intervention can reverse paclitaxel resistance to some extent.
