益生菌DNA干预对变应性鼻炎疗效的动物实验研究

【摘要】:目的 益生菌DNA滴鼻干预对卵清蛋白(OVA)诱发的变应性鼻炎疗效的动物模型研究,探讨小鼠鼻黏膜中调节性T细胞(Treg细胞)及血清Th1/ Th2细胞因子水平。 方法 培养益生菌,提取DNA,取6-8周Balb/c小鼠随机分为益生菌DNA滴鼻组及生理盐水对照组,每组15只。益生菌滴鼻组:30%的卵清蛋白(OVA)生理盐水溶液,每毫升加入AL(OH)330 mg腹腔注射大鼠,隔日1次,每次1 ml,7次后用5%的OVA液滴鼻,每侧l0ul,每天1次,共7次,致敏的同时取已提取的DNA液滴鼻,每侧5ul(25ug/ml),每天一次。对照组:OVA造模方法同益生菌滴鼻组,但以生理盐水代替益生菌DNA液滴鼻。采用ELISA法检测各组血清IgE及IL-4,IL-10,IFN-γ,IFN-γ/ IL-4水平。分析鼻黏膜中Foxp3阳性细胞数,并对鼻黏膜行组织形态学观察。组间用t检验。 结果 1.益生菌DNA滴鼻组IgE显著低于对照组(p0.05), 2.益生菌DNA滴鼻组IL-10,IFN-γ,IL-4均低于对照组(p0.05),而IFN-γ/IL-4高于对照组(p0.05)。 3.益生菌DNA滴鼻组鼻粘膜中Foxp-3阳性细胞数显著高于对照组(p0.05)。 4.益生菌DNA滴鼻组鼻粘膜较对照组出现明显的形态学改变。 结论 1.益生菌DNA滴鼻干预通过降低动物模型IgE水平,缓解Ⅰ型变态反应性。 2.益生菌DNA滴鼻通过调整模型动物Th1/Th2细胞因子平衡,,抑制Th2细胞反应,对变应性鼻炎起一定防治作用。 3.益生菌DNA滴鼻早期干预可提高动物模型鼻粘膜Foxp3阳性细胞数抑制小鼠AR的进展。
[Abstract]:Objective to investigate the effects of probiotic DNA nasal drip on allergic rhinitis induced by ovalbumin (OVA) and to investigate the levels of regulatory T cells (Treg cells) and serum Th1/ Th2 cytokines in mouse nasal mucosa. Methods probiotics were cultured and Balb/c mice were randomly divided into probiotic DNA nasal dropping group and normal saline control group for 6 to 8 weeks with 15 mice in each group. Probiotic nasal drip group: 30% ovalbumin (OVA) saline solution was injected intraperitoneally into rats with AL (OH) 330 mg per milliliter. Once a second day, 1 ml,7 per day, 5% OVA solution was used on each side of the nose, 10% per side, once a day, for 7 times. At the same time, the extracted DNA solution was nasal drops, 5ul (25ug/ml) on each side, once a day. The control group was treated with probiotic nasal drip, but normal saline was used instead of probiotic DNA solution. Serum IgE and IL-4,IL-10,IFN- 纬 / IL-4 levels were detected by ELISA assay. The number of Foxp3 positive cells in nasal mucosa was analyzed and the histomorphology of nasal mucosa was observed. T test was used between groups. Result 1. The IgE of probiotic DNA nasal drip group was significantly lower than that of control group (p0.05). The levels of IL-10,IFN- 纬 -IL-4 in probiotic DNA nasal drip group were lower than those in control group (p0.05), while IFN- 纬 -IL-4 was higher than that in control group (p0.05). The number of Foxp-3 positive cells in nasal mucosa of probiotic DNA dropping group was significantly higher than that of control group (p0.05). The nasal mucosa of the probiotic DNA nasal drip group showed obvious morphologic changes compared with the control group. Conclusion 1. Probiotic DNA nasal drip alleviates type 鈪
