8-OHdG和VEGF在原发性翼状胬肉组织中的表达及意义

【摘要】:目的通过检测8-羟基脱氧鸟苷(8-OHd G)及血管内皮生长因子(VEGF)在原发性翼状胬肉和正常结膜组织中的表达,探讨DNA氧化损伤及新生血管形成在原发性翼状胬肉发病机制中的作用。方法收集手术切除的原发性翼状胬肉组织标本35例(35眼),并收集术眼颞上方正常球结膜标本5例(5眼)作对照。分别采用免疫组织化学法和实时荧光定量PCR技术检测翼状胬肉和结膜组织标本中8-OHd G及VEGF的表达,分析比较两种因子在不同组织间的表达差异及表达规律。结果1.8-OHd G在翼状胬肉和结膜组织标本中的表达差异及规律在35例翼状胬肉组织中有24例呈阳性表达,阳性表达率为68.6%,而正常结膜组织中无8-OHd G的表达,其阳性表达率的差异具有显著统计学意义(P=0.007)。8-OHd G的阳性表达位于胬肉组织上皮细胞的细胞核、呈棕黄色着染,细胞质无染色。上皮下的纤维血管组织及正常结膜组织无表达。2.VEGF在翼状胬肉和结膜组织标本中的表达差异及规律(1)VEGF m RNA在翼状胬肉各部位及正常球结膜(1.18323±0.702762)中均有表达,且自翼状胬肉头部(0.99526±0.525440)、颈部(1.38062±0.715145)至体部(2.41824±0.906681)表达量呈逐渐增高趋势。(2)VEGF m RNA在翼状胬肉体部的表达量明显高于头部和颈部,表达的差异均具有显著的统计学意义(P0.001)。(3)翼状胬肉体部VEGF的表达较正常结膜明显增高,差异具有显著统计学意义(P=0.0020.01);但头部、颈部与正常结膜三者之间的表达差异无明显统计学意义(P0.05)。结论1.8-OHd G及VEGF在原发性翼状胬肉组织中的表达均比正常结膜中表达明显增高,提示DNA氧化应激损伤及新生血管形成在原发性翼状胬肉发病机制中发挥重要作用。2.VEGF在原发性翼状胬肉各部位的表达自头部至体部呈逐渐增高趋势,提示翼状胬肉的新生血管过度增生位于胬肉体部。
[Abstract]:Objective to detect the expression of 8-hydroxydeoxyguanosine (8-OHd G) and vascular endothelial growth factor (VEGF) in primary pterygium and normal conjunctiva. To investigate the role of DNA oxidative injury and angiogenesis in the pathogenesis of primary pterygium. Methods 35 cases (35 eyes) of primary pterygium were collected and 5 cases (5 eyes) of normal bulbar conjunctiva were collected as control. The expression of 8-OHd G and VEGF in pterygium and conjunctiva was detected by immunohistochemistry and real-time fluorescence quantitative PCR. Results the expression of 1.8-OHd G in pterygium and conjunctiva tissues was different and regular. 24 cases of pterygium showed positive expression in 35 cases of pterygium, the positive rate was 68.6%, but there was no expression of 8-OHd G in normal conjunctiva. The positive expression rate of 8-OHd G was located in the nucleus of pterygium epithelium cells and was stained with brown color and no staining in cytoplasm. The expression of 2.VEGF in pterygium and conjunctiva tissues was different (1) VEGF m RNA was expressed in all parts of pterygium and normal bulbar conjunctiva (1.18323 卤0. 0). 702762). The expression of pterygium was increased from head (0.99526 卤0.525440), neck (1.38062 卤0.715145) to body (2.41824 卤0.906681). (2) the expression of pterygium in pterygium was significantly higher than that in head and neck. The results suggest that oxidative stress injury and angiogenesis of DNA play an important role in the pathogenesis of primary pterygium. The expression of 2.VEGF in all parts of primary pterygium is increasing gradually from head to body. The results suggest that the hyperplastic neovascularization of pterygium is located in pterygium.
