转移抑素对鼻咽癌高转移能力细胞株SUNE-1-5-8F转移侵袭能力的影响

【摘要】:目的:研究并探讨转移抑素(metastin)对鼻咽癌高转移能力细胞株SUNE-1-5-8F转移侵袭能力的影响。方法:(1)通过Transwell体外侵袭趋化实验技术,测定在不同浓度(102nM,103nM,104nM)的转移抑素干预前后SUNE-1-5-8F细胞转移及侵袭细胞数目,探究其对SUNE-1-5-8F细胞转移及侵袭能力的影响。(2)利用BALB/c-nu裸鼠肺转移模型,40只雌性裸鼠随机分为转移抑素组及对照组,每组各20只,两组均在腋窝下注射SUNE-1-5-8F细胞悬液2×106/ml,其中转移抑素组于注射细胞后第二天按1.17mg/kg(转移抑素量/裸鼠体重)腹腔注射转移抑素,研究转移抑素对裸鼠移植瘤的抑瘤率、转移抑制率及生命延长率的影响;采用免疫组化法研究裸鼠肺转移灶kisspeptin指标表达情况。结果:(1)Transwell体外侵袭趋化实验中转移抑素对于鼻咽癌高转移能力细胞SUNE-1-5-8F的转移能力及侵袭能力均具有显著的抑制作用(P0.05),且随着转移抑素浓度的提升与其抑制转移侵袭能力呈正相关(P0.05)。(2)BALB/c-nu裸鼠肺转移模型实验中转移抑素对肿瘤的生长即抑瘤率无明显的影响(P0.05),但可以有效的降低肺转移数,提升其转移抑制率(P0.05),且进一步提升了小鼠的生命延长率(P0.05)。免疫组化结果显示转移抑素组kisspeptin较对照组高表达。结论:(1)在Transwell体外侵袭趋化实验中,转移抑素能够明显抑制鼻咽癌高转移能力细胞株SUNE-1-5-8F的转移侵袭能力,且转移抑素浓度与其抑制能力呈正相关。(2)在BALB/c-nu裸鼠肺转移模型实验中,转移抑素能够有效降低肺转移率,从而延长裸鼠生命,但无法抑制肿瘤的生长。
[Abstract]:Objective: to investigate the effect of metastatic (metastin) on metastasis and invasion of NPC cell line SUNE-1-5-8F. Methods: (1) Transwell invasion chemotactic assay was used to determine the number of SUNE-1-5-8F cell metastasis and invasion cells before and after treatment with different concentrations (102nM) of 103nMU 104nM. To investigate the effect of SUNE-1-5-8F on the metastasis and invasion of SUNE-1-5-8F cells. (2) using the lung metastasis model of BALB/c-nu nude mice, 40 female nude mice were randomly divided into metastatic statin group and control group with 20 in each group. SUNE-1-5-8F cell suspension was injected into the axilla of both groups (2 脳 10 6 / ml). The metastatic statin group was injected intraperitoneally with 1.17mg/kg on the second day after injection of the cells. To study the effect of metastasis statin on tumor inhibition rate, metastasis inhibition rate and life prolongation rate of xenograft tumor in nude mice. The expression of kisspeptin in lung metastases in nude mice was studied by immunohistochemical method. Results: (1) in vitro invasion and chemotactic assay of Transwell, metastatic statin significantly inhibited the metastasis and invasion of SUNE-1-5-8F cells with high metastatic ability of nasopharyngeal carcinoma (P0.05). And with the increase of the concentration of metastasis, there was a positive correlation between the inhibition of metastasis and invasion ability (P0.05). (2) in the lung metastasis model of BALB/c-nu nude mice. There was no significant effect of metastasis on tumor growth and tumor inhibition rate (P0.05). But can effectively reduce the number of lung metastasis, enhance its metastasis inhibition rate (P0.05), and further enhance the survival rate of mice (P0.05). Immunohistochemical results showed that the expression of kisspeptin in the metastatic statin group was higher than that in the control group. Conclusion: (1) Metastatin can significantly inhibit the metastasis and invasion ability of nasopharyngeal carcinoma cell line SUNE-1-5-8F in vitro invasion and chemotaxis assay. The concentration of metastatic statin was positively correlated with its inhibitory ability. (2) in the lung metastasis model of BALB/c-nu nude mice, metastatic statin could effectively reduce the rate of lung metastasis and prolong the life of nude mice, but could not inhibit the growth of tumor.
