慢性间歇性缺氧对大鼠食管及其IL-6,hs-CRP表达的影响

【摘要】:目的: 制作常压下慢性间歇性缺氧的大鼠模型,以探讨CIH与胃食管反流病的关系及CIH对食管白细胞介素-6(Interleukin-6,IL-6),超敏c-反应蛋白(high-sensitivity C-reactive protein,hs-CRP)表达的影响。 方法: 将24只成年雄性SD大鼠随机分成3组,即空白对照组(unhandledcontrol group,UC组,按夜间进食、水,而白天禁食、水,不限制活动,不加任何人工低氧干预的方案饲养4周),慢性间歇性缺氧组(chronic intermittent hypoxia group,CIH组,按UC组方法喂养,连续4周给予每天白天8h的间歇性缺氧)和复氧组(Removal of hypoxiagroup,RH组,按CIH方法喂养4周后再按UC组方法饲养4周),所有大鼠均在按规定完成实验后测定各组食管下段PH值,再在肉眼及光镜下观察食管病理变化,,用酶联免疫法检测食管组织IL-6及hs-CRP水平。 结果: 1.CIH组食管下段PH(potential of hydrogen)值较UC组明显下降(p0.05),RH组与UC组无明显差异; 2. CIH组食管发生明显炎症改变,UC组和RH组未发现明显的炎症性改变; 3. CIH组IL-6及hs-CRP的表达明显增高(P0.01),RH组与UC组比较无统计学差异(p0.05)。 结论: CIH可使大鼠食管PH值下降,食管炎性改变明显,IL-6及hs-CRP表达升高,复氧可使其恢复。
[Abstract]:Objective: to study the relationship between CIH and gastroesophageal reflux disease (GERD) and the effect of CIH on esophageal interleukin-6 (Interleukin-6,IL-6) and high-sensitivity C-reactive protein, in rats with chronic intermittent hypoxia under normal pressure. Hs-CRP) expression. Methods: Twenty-four adult male SD rats were randomly divided into three groups: control group (unhandledcontrol group,UC group, fed with night-time food, water, daytime fasting, water, no restriction of activity, and no artificial hypoxic intervention for 4 weeks). The chronic intermittent hypoxia group (chronic intermittent hypoxia group,CIH group) was fed with UC for 4 weeks, and the reoxygenation group (Removal of hypoxiagroup,RH group) and the reoxygenation group (Removal of hypoxiagroup,RH group) were fed with CIH method for 4 weeks and then fed with UC group for 4 weeks). The PH values of lower esophagus in all groups were measured after completion of the experiment. The pathological changes of esophagus were observed with naked eye and light microscope. The levels of IL-6 and hs-CRP in esophageal tissue were detected by enzyme-linked immunosorbent assay (Elisa). Results: the PH (potential of hydrogen) value of lower esophagus in 1.CIH group was significantly lower than that in UC group (p0.05), RH group and UC group had no significant difference; 2. In CIH group, there were obvious inflammatory changes in esophagus, but no obvious inflammatory changes were found in UC group and RH group. The expression of IL-6 and hs-CRP in CIH group was significantly higher than that in UC group (P 0.01). Conclusion: CIH can decrease the value of PH in rat esophagus, increase the expression of IL-6 and hs-CRP, and reoxygenate can make it recover.
