原花青素对人眼小梁细胞抗氧化损伤的实验研究

【摘要】:目的: 研究原花青素(Procyanidins,PC)对过氧化氢(Hydrogen peroxide)处理的人眼小梁细胞(Human Trabecular Meshwork Cells,HTMC)抗氧化损伤的保护作用,为青光眼的临床治疗提供实验依据。 方法: 将正常人眼小梁细胞进行细胞传代后随机分为5组。未处理组:正常培养的HTMC,对照组:正常培养的HTMC+过氧化氢;实验组:正常培养的HTMC+过氧化氢+PC(PC浓度分别为0.02mg/ml、0.05mg/ml、0.10mg/ml)。应用实时荧光定量PCR方法检测线粒体复合物I mRNA(complex Ⅰ mRNA)的表达,流式细胞术检测细胞凋亡率,Western blot检测细胞色素C(cytochrome C,CytC)的释放。 结果: 1.与对照组相比,各实验组线粒体complex Ⅰ mRNA表达均增加,CytC释放均减少,细胞凋亡率均降低,差异均有统计学意义(P0.01)。2.不同浓度PC组间随PC浓度增加complex Ⅰ mRNA表达增加,CytC释放减少,细胞凋亡率降低,各浓度组间差异有统计学意义(P0.01)。3.与未处理组相比,各PC组细胞凋亡率差异均有统计学意义(P0.01),,0.02mg/ml和0.05mg/mlPC组complexI mRNA表达、CytC释放差异均有统计学意义(P0.01),而0.10mg/mlPC组差异均无统计学意义(P0.05)。 结论: 外源性PC可以增加氧化损伤人眼小梁细胞线粒体complex Ⅰ mRNA的表达,减少CytC的释放,降低细胞凋亡率,有较强的抗氧化作用,并在一定浓度范围内随着浓度的增加其抗氧化作用逐渐增强,PC在青光眼的临床治疗中的作用值得进一步研究。
