甲状腺相关眼病患者外周血单个核细胞中微小RNA-146a的异常表达及意义

【摘要】:目的探讨微小RNA-146a (microRNA-146a, miRNA-146a, miR-146a)在甲状腺相关眼病(TAO)患者外周血单个核细胞中的表达情况及临床意义,为TAO的治疗提供新思路。 方法收集TAO患者、Gaves病(GD)不伴眼病患者和正常人对照者空腹静脉全血标本各20例,分离各组患者外周血单个核细胞(PBMCs),用TRIzol提取PBMCs的总miRNA,再以颈环结构的引物逆转录出相应的DNA,然后以miR-146a的PCR引物进行实时荧光定量PCR(qRT-PCR)检测各组标本PBMCs中miR-146a的表达情况,电泳观察结果. 结果与正常人相比,TAO患者和GD不伴眼病患者PBMCs中miR-146a相对表达量明显降低(P0.05),下调率分别为98.02%和67.60%;较GD不伴眼病患者,miR-146a在TAO患者PBMCs中相对表达量更低(P0.05),下调率差值为30.40%。 结论1.基于SYBR Green Ⅰ染料荧光分析的茎环qRT-PCR方法检测miR-146a表达的特异性良好; 2、miR-146a的表达下调可能参与了TAO的发病机制,为进一步研究微小RNA在TAO发病机制中的作用打下基础。
[Abstract]:Objective To study the expression and clinical significance of microRNA-146a (microRNA-146a, miRNA-146a, miR-146a) in peripheral blood mononuclear cells of patients with thyroid-associated ophthalmopathy (TAO) and to provide a new thought for the treatment of TAO. Methods 20 patients with TAO, Gaves disease (GD) and normal controls were collected, and the peripheral blood mononuclear cells (PBMCs) were isolated and the total miRNAs of PBMCs were extracted with TRIzol. A. The expression of miR-146a in PBMCs of each group was detected by real-time fluorescence quantitative PCR (qRT-PCR) using the PCR primer of miR-146a. Results Compared with normal subjects, the relative expression of miR-146a in patients with TAO and GD was significantly lower than that of normal subjects (P0.05). The down-regulation rate was 98.02% and 67.60%, respectively. The relative expression of miR-146a in PBMCs of patients with GD was lower (P0.05), and the difference of down-regulation was 30. 40%. Conclusion 1. The expression of miR-146a is detected by the stem-loop qRT-PCR method based on the fluorescence analysis of SYBR Green I dye. Good specificity; the down-regulation of miR-146a may be involved in the pathogenesis of TAO, in order to further study the mechanism of small RNA in the pathogenesis of TAO
