姜黄素对核转录因子-κB表达的影响及小鼠角膜紫外线照射损伤的抑制作用

【摘要】:目的探讨姜黄素对紫外线A(ultraviolet A,UVA)照射小鼠角膜损伤的抑制作用。方法取C57 BL/6小鼠30只,随机分为3组,每组各10只;UVA照射组双眼接受位于前上方的UVA照射,波长为320~400 nm,照射强度为0.05 W·cm~(-2),照射距离固定,照射时间24 h。姜黄素干预组在UVA照射前3 d给予小鼠姜黄素30 mg·kg~(-1)腹腔内注射,并持续至照射后48 h。对照组不做任何处理。利用裂隙灯观察角膜病变并对角膜混浊进行分级;照射后48 h取小鼠角膜组织,使用电泳迁移率实验检测核转录因子-κB(nuclear factor-κB,NF-κB)的表达。结果照射后48 h,UVA照射组角膜混浊临床评分(3.10±0.74)均高于对照组(0)及姜黄素干预组(0.35±0.24)。两两比较发现对照组与姜黄素干预组间角膜混浊临床评分差异无统计学意义(P0.05);UVA照射组与对照组以及姜黄素干预组间差异均有统计学意义(均为P0.05)。照射后48 h,角膜组织NF-κB表达水平对照组(1.25±0.13)与姜黄素干预组(1.58±0.34)均较低,UVA照射组(3.98±0.58)明显升高。两两比较发现对照组与姜黄素干预组角膜组织NF-κB表达水平差异无统计学意义(P0.05);UVA照射组与对照组以及姜黄素干预组间差异均有统计学意义(均为P0.05)。结论姜黄素可以减轻UVA照射对小鼠角膜造成的损伤,其作用可能是通过抑制角膜组织内NF-κB表达实现的。
[Abstract]:Objective to investigate the inhibitory effect of curcumin on corneal injury induced by ultraviolet A (UVA) irradiation in mice. Methods Thirty C57 BL/6 mice were randomly divided into three groups with 10 mice in each group. In the UVA group, the eyes were exposed to the anterior-superior UVA irradiation. The irradiation intensity was 0.05 W cm~ (- 2) at the wavelength of 320 渭 400 nm,. The irradiation distance was fixed and the irradiation time was 24 h. Curcumin intervention group was injected intraperitoneally with curcumin 30 mg kg~ (- 1) 3 days before UVA irradiation and lasted until 48 hours after irradiation. The control group did not do any treatment. The keratopathy was observed by slit lamp and the corneal opacity was graded, and the expression of nuclear factor-魏 B (nuclear factor- kappa B (NF- 魏 B) was detected by electrophoretic mobility shift assay (EMSA) 48 h after irradiation. Results 48 hours after irradiation, the clinical score of corneal opacification in UVA irradiation group (3.10 卤0.74) was higher than that in control group (0) and curcumin intervention group (0.35 卤0.24). There was no significant difference in the clinical score of corneal opacification between the control group and curcumin intervention group (P0.05) between the); UVA irradiation group and the control group and curcumin intervention group (P0.05). 48 hours after irradiation, the expression level of NF- 魏 B in corneal tissue was lower in control group (1.25 卤0.13) and curcumin intervention group (1.58 卤0.34), while that in UVA group was significantly higher than that in control group (3.98 卤0.58). Pairwise comparison showed that there was no significant difference in the expression of NF- 魏 B between the control group and curcumin intervention group (P0.05) between the); UVA irradiation group and the control group and curcumin intervention group (P0.05). Conclusion curcumin can reduce the corneal damage induced by UVA irradiation in mice, which may be achieved by inhibiting the expression of NF- 魏 B in corneal tissue.
