B淋巴细胞活化因子促进幽门螺杆菌诱导的十二指肠溃疡患者Th17细胞分化

【摘要】:目的:分析幽门螺杆菌诱发十二指肠溃疡患者血清中B淋巴细胞活化因子(BAFF)表达情况,探讨BAFF对Th17细胞分化的影响。方法:ELISA法测患者血清中BAFF与IL-17浓度;MTT法测T细胞与幽门螺杆菌共培养后存活率;流式细胞术测BAFF-R表达;RT-PCR、Western blot、ELISA法测RORγt和RORα及IL-17、IL-21、IL-22的m RNA和蛋白表达。结果:与对照相比,幽门螺杆菌阳性患者血清中BAFF与IL-17表达水平显著增高(P0.05),两者表达水平呈正相关(r=0.87,P0.01)。BAFF处理后,幽门螺杆菌诱导的T细胞生存活力较对照增强,BAFF-R表达增加,Th17细胞标志性转录因子RORγt和RORα及细胞因子IL-17、IL-21、IL-22的m RNA和蛋白表达水平均明显高于对照(P0.05)。结论:幽门螺杆菌能引起十二指肠溃疡患者体内BAFF高表达,进而促进Th17细胞分化。
[Abstract]:Objective: to analyze the expression of B lymphocyte activating factor (BAFF) in serum of patients with duodenal ulcer induced by Helicobacter pylori. To investigate the effect of BAFF on the differentiation of Th17 cells. Methods the survival rate of T cells and Helicobacter pylori co-cultured was measured by BAFF and IL-17 concentration in serum of patients. Flow cytometry was used to detect the expression of ROR 纬 t and ROR 伪, and the m RNA and protein of ROR 伪 and IL-17 IL-21, IL-22. Results: compared with the control group, the expression of BAFF and IL-17 in the serum of patients with Helicobacter pylori positive was significantly higher than that in the control group. The expression of BAFF and IL-17 in serum of patients with Helicobacter pylori positive was significantly higher than that of the control group. The survival activity of T cells induced by Helicobacter pylori was significantly higher than that of the control group. The expression levels of ROR 纬 t and ROR 伪, as well as the expression levels of m RNA and protein of IL-17, IL-21, IL-22 in Th17 cells were significantly higher than those in the control group P0.050.Conclusion: Helicobacter pylori is more potent than the control group. Can cause high expression of BAFF in patients with duodenal ulcer. And then promote the differentiation of Th17 cells.
