类泛素缀和酶9在肝纤维化中的作用及机制

【摘要】:目的研究类泛素缀和酶9(UBC9)在肝星状细胞的激活及肝纤维化中的作用机制。方法通过蛋白印记法(Western blot)检测UBC9在不同浓度的转化生长因子-β1(TGF-β1)刺激下的肝星状细胞LX-2中的表达情况;构建UBC9质粒干扰片段(UBC9-shRNA),采用脂质体介导转染LX-2,同时设置无义序列组和空白对照组,实时荧光定量PCR检测转染后UBC9 mRNA的表达;Western blot检测UBC9在蛋白水平的表达;Western blot检测下调UBC9对α-SMA、Collagen Ⅰ及磷酸化的Smad3表达的影响。CCK8检测转染后细胞增殖能力。结果 UBC9在TGF-β1因子刺激下的肝星状细胞LX-2表达增加,一定剂量下呈浓度依赖性。转染干扰片段UBC9-shRNA后,激活的LX-2细胞增殖减弱,α-SMA和Collagen Ⅰ蛋白的表达减少,而且下调UBC9也能抑制磷酸化的Smad3的表达。结论下调UBC9可以通过抑制TGF-β1/Smad3信号通路进而影响肝星状细胞LX-2的激活,这些都提示UBC9在肝纤维化过程具有重要作用,为肝纤维化治疗提供一些新的理论依据。
[Abstract]:Objective to study the role of UBC9 and UBC9 in the activation and fibrosis of hepatic stellate cells. Methods the expression of UBC9 in hepatic stellate cells stimulated by TGF- 尾 1 at different concentrations was detected by Western blot. UBC9 plasmid interference fragment UBC9-shRNA was constructed and transfected into LX-2 by liposome-mediated transfection. At the same time, nonsense sequence group and blank control group were set up. The expression of UBC9 mRNA after transfection was detected by real-time fluorescence quantitative PCR.
