视黄醇结合蛋白4在2型糖尿病组和血糖正常组胆囊组织中的表达差异

【摘要】：目的探讨视黄醇结合蛋白4(RBP4)的蛋白表达量及RBP4 m RNA表达量在胆囊结石合并2型糖尿病组和未合并糖尿病组的胆囊组织中的差异。方法首先应用免疫组化检测30例病例组与30例对照组胆囊组织上RBP4的表达情况,然后用Western blot技术检测胆囊黏膜组织上RBP4的表达量,并与相关临床资料结合进行统计学分析。结果免疫组化检测到RBP4在病例组胆囊组织中的表达强度明显高于对照组(P0.001),女性表达强度明显高于男性(P0.001),腹型肥胖者表达强度明显高于非腹型肥胖者(P0.001);Western blot结果显示:RBP4在病例组的胆囊黏膜组织上的蛋白相对表达量为(2.91±0.86),对照组为(2.17±0.73),差异有统计学意义(P0.001),病例组RBP4在胆囊黏膜组织中的蛋白表达量明显高于对照组。结论通过免疫组化和Western blot实验,在分子生物学的层面上证明了2型糖尿病组胆囊组织的RBP4表达水平高于对照组,同时女性RBP4的表达强度高于男性,腹型肥胖者RBP4的表达强度高于非腹型肥胖者。
[Abstract]:Objective to investigate the expression of retinol binding protein 4 (RBP4) and the expression of RBP4 m RNA in gallbladder tissues of type 2 diabetes patients with gallstone and those without diabetes mellitus. Methods the expression of RBP4 in gallbladder tissues of 30 cases and 30 cases of control group was detected by immunohistochemistry, then the expression of RBP4 in gallbladder mucosa was detected by Western blot technique. Results Immunohistochemical staining showed that the expression of RBP4 in gallbladder tissue of the case group was significantly higher than that of the control group (P 0.001), the expression intensity of female was significantly higher than that of male (P 0.001), and the expression intensity of abdominal obesity was significantly higher than that of non-abdominal obesity (P 0.001). Western blot results showed that the expression of RBP4 in gallbladder tissue was significantly higher than that in control group. The relative expression of RBP4 in gallbladder mucosa was 2.91 卤0.86 in the case group and 2.17 卤0.73 in the control group. The difference was statistically significant (P 0.001). The expression of RBP4 in the gallbladder mucosa in the case group was significantly higher than that in the control group. Conclusion on the level of molecular biology, the expression of RBP4 in gallbladder tissue of type 2 diabetes mellitus is higher than that of control group, and the expression of RBP4 in women is higher than that in men. The expression of RBP4 in abdominal obesity was higher than that in non-abdominal obesity.
