乌司他丁对大鼠重症急性胰腺炎肠黏膜屏障功能的影响

【摘要】:目的探讨乌司他丁(UTI)对重症急性胰腺炎(SAP)大鼠肠黏膜屏障的影响及其作用机制。方法 SD大鼠60只分成对照组、假手术(SO)组、SAP组和UTI组。制模后每隔8 h进行腹腔内注射UTI(浓度1.5万IU/kg)作为UTI组。各治疗组检测制模后8、24、72 h血浆内毒素水平,通过RT-PCR法和免疫组化法对肠黏膜Toll样受体(TLR)4表达进行检测,并在电镜下观察肠黏膜组织。结果对照组肠黏膜未见TLR4表达。与对照组相比,SAP组在制模后8 h血浆内毒素和肠黏膜TLR4表达即出现升高,并随时间的延长而继续进行性增高。血浆内毒素与肠黏膜TLR4二者明显相关。UTI组同时点内毒素、TLR4低于SAP组,电镜下检查发现病理损伤也同时减轻,24 h时最为明显。结论 UTI能有效保护SAP发生时的肠屏障功能,其保护机制可能与肠黏膜TLR4的表达下调有关。
[Abstract]:Objective to investigate the effect and mechanism of ulinastatin (UTI) on intestinal mucosal barrier in rats with severe acute pancreatitis (SAP). Methods 60 SD rats were divided into control group, sham operation group and UTI group. The UTI group was treated by intraperitoneal injection of UTI (15000 IU / kg) every 8 hours. The plasma endotoxin level was measured at 824 hours after model making in each treatment group. The expression of Toll like receptor TLR4 in intestinal mucosa was detected by RT-PCR and immunohistochemistry, and the intestinal mucosal tissue was observed under electron microscope. Results there was no TLR4 expression in intestinal mucosa of control group. Compared with the control group, plasma endotoxin and intestinal mucosal TLR4 expression increased at 8 h after model making, and continued to increase progressively with the prolongation of time. There was a significant correlation between plasma endotoxin and intestinal mucosal TLR4. TLR4 in UTI group was lower than that in SAP group at the same time. Conclusion UTI can effectively protect the intestinal barrier function during the occurrence of SAP, and its protective mechanism may be related to the down-regulation of TLR4 expression in intestinal mucosa.
