中药高位保留灌肠对STC大鼠结肠ICC与血清VIP的影响研究

【摘要】:目的:观察通腑润肠汤高位保留灌肠治疗慢传输型便秘(STC)模型大鼠的效果及对结肠Cajal间质细胞(ICC)、血管活性肠肽(VIP)的影响,探讨通腑润肠汤高位保留灌肠治疗STC的效果及作用机制。方法:将60只SD大鼠随机分为A组(空白对照组)、B组(便秘模型组)、C组(莫沙必利组)、D组(中药高位保留灌肠低剂量组)、E组(中药高位保留灌肠中剂量组)、F组(中药高位保留灌肠高剂量组)。造模期间用复方地芬诺酯对B组、C组、D组、E组、F组灌胃造模,用生理盐水对A组灌胃。造模成功后,A组、B组用生理盐水灌胃,剩余各组用对应药物治疗。观察各组大鼠造模前、造模成功后及治疗完成后的一般情况、24h内的粪便粒数及粪便的含水率,治疗结束后检测SD大鼠的结肠炭末推进率、血清VIP浓度、结肠ICC数量。结果:(1)在排便粒数上,C、D、E、F组和B组比较的p值均为0.000,P0.05;E组和C组、D组比较的p值均为0.001,p0.05;F组和C组、D组比较的p值均为0.001,p0.05;C组、D组比较比较的p值0.930,p0.05;E组、F组比较的p值为0.784,p0.05。(2)在粪便含水率上,B组和C组比较的p值为0.820,p0.05;B和D、E、F组比较的p值为依次为0.002、0.001、0.001,p0.05;D组和E、F组比较的p值依次为0.721、0.752,p0.05;E组和F组比较的p值为0.960,p0.05;(3)在结肠炭末推进率上,B组和C、D、E、F组比较的p值都为0.000,p0.05;E组和C、D组比较的p值都为0.000,p0.05;F组和C、D组比较的p值都为0.000,p0.05;C组和D组比较的p值为0.710,p0.05;E组和F组比较的p值为0.964,p0.05。(4)在影响VIP浓度上,C、D、E、F组和B组比较的p值都依次为0.000、0.001、0.000、0.000,P0.05;而C组和D、E、F组比较的p值依次为0.593、0.954、0.643;D组和E、F组比较的p值依次为0.642、0.933;E组和F组比较的p值为0.694,p0.05。(5)在影响ICC数量上,C、D组与B组比较的p值依次为0.311、0.106,P0.05;E、F组与B组比较的p值依次0.006、0.020,p0.05。结论:通腑润肠汤高位保留灌肠通过增加结肠ICC数量、血清VIP浓度来增加排便量、提高粪便含水率、促进肠道传输以改善便秘症状。通腑润肠汤高位保留灌肠中、高剂量疗效相当,均较通腑润肠汤高位保留灌肠低剂量及莫沙必利治疗效果好。
[Abstract]:Objective: to observe the effect of Tongfu Runchang decoction high position retention enema on slow transit constipation (STC) model rats and the effect of Tongfu Runchang decoction on (ICC), vasoactive intestinal peptide (VIP) in interstitial cells of colon Cajal. To explore the effect and mechanism of Tongfu Runchang decoction (Tongfu Runchang decoction) in the treatment of STC with high retention enema. Methods: sixty Sprague-Dawley rats were randomly divided into two groups: group A (control group), group B (constipation model group), group C (mosapride group), group D (low dose group of high retention enema of traditional Chinese medicine) and group E (middle dose group of high retention enema of traditional Chinese medicine). Chinese medicine high retention enema high dose group). During the course of modeling, compound diphenoxylate was used to make models in group B, group C, group D, group C, group E, group F, and group A, with normal saline. The rats in group A were given normal saline and the rest were treated with corresponding drugs. The number of fecal particles and the fecal moisture content were observed before, after and after treatment. The colon carbon propelling rate, serum VIP concentration and colonic ICC content of SD rats were measured after treatment. Results: (1) the p values of group F and group B were both 0.000, P0.05E and C, respectively. The p values of group F and group C were both 0.001p0.05F and 0.001p0.05C, respectively. The p values of group D and group D were 0.930 and 0.05g respectively. (2) the p values of group B and group C were 0.820, p0.05B and group F, respectively. The p values of group D and group E were 0.7210.752p0.05E and 0.960p0.05E, respectively; (3) the rate of propelling of colon carbon in group B and group C were 0.960p0.05; (3) in group B and group C, the p values of group B and group C were 0.960p0.05; (3) the p values of group B and group C were respectively 0.7210.752p0.05E and 0.960p0.05e; (3) the p values of group B and group C were higher than those of group F and group B (P = 0.960p0.05) respectively; (3) the p values of group B and group C were 0.960p0.05 respectively. The p values of group E and C D were both 0.000g / d and 0.000g / d, respectively. The p values of group C and group D were both 0.000g / g and 0.000g / d, respectively. The p values of group C and group D were 0.710g / p0.05E and 0.964g / 0.05p respectively. (4) the difference of VIP concentration between group F and group B was found to be higher than that of group C and group D (P = 0.605), respectively. Conclusion: Tongfu Runchang decoction high retention enema can increase fecal excretion, increase fecal moisture and improve intestinal transport to improve constipation symptoms by increasing the number of colon ICC and serum VIP concentration. In the high retention enema of Tongfu Runchang decoction, the effect of high dose retention enema was equal, which was better than that of Tongfu Runchang decoction high retention enema low dose and mosapride.
