龙血竭联合鱼腥草对UC模型大鼠OFR影响的实验研究

【摘要】:目的:观察龙血竭联合鱼腥草对UC模型大鼠疾病活动指数、结肠黏膜大体形态、病理组织学改变,并检测UC模型大鼠血清超氧化物歧化酶(SOD)与丙二醛(MDA)的变化,验证鱼腥草及龙血竭对UC的治疗作用,探讨二者联合应用是否对UC的治疗有协同作用及其作用机理,从而为UC规范化治疗及用药提供依据。方法:将SD大鼠随机分为正常对照组和模型组。模型组予以2,4,6-三硝基苯磺酸加乙醇灌肠造模。当UC模型造模成功后,将模型组大鼠随机分为龙血竭组,鱼腥草组,龙血竭合鱼腥草组低、中、高剂量组,柳氮磺吡啶(SASP)组及模型对照组。分别给药灌胃两周后,心脏采血进行血清SOD、MDA检测,剖腹留取病变结肠做病理切片。结果:药物灌胃后,各灌胃组大鼠体重均有增加,龙血竭合鱼腥草高、中剂量组两者之间比较,差异无统计学意义(P0.05);龙血竭合鱼腥草高、中剂量组与SASP组、龙血竭组、鱼腥草组相比,体重增加量更大,差异具有统计学意义(P0.05)。各灌胃组大鼠结肠粘膜充血水肿,糜烂及溃疡等症状均有改善,龙血竭合鱼腥草高、中剂量组、龙血竭组、鱼腥草组四组之间比较,差异无统计学意义(P0.05);龙血竭合鱼腥草高、中剂量组、龙血竭组、鱼腥草组与SASP组相比,结肠黏膜损伤指数更低,差异具有统计学意义(P0.05)。各灌胃组血清中SOD浓度均有升高,血清中MDA浓度均有降低,在幅度上龙血竭合鱼腥草高、中剂量组与SASP组、鱼腥草组、龙血竭组相比,差异具有统计学意义(P0.05)。结论:本实验研究表明龙血竭合鱼腥草高、中剂量组能够改善结肠粘膜充血水肿、糜烂出血、溃疡等症状,提高血清SOD活性、降低MDA含量,且优于SASP组、鱼腥草组、龙血竭组,提示龙血竭合鱼腥草中、高剂量组对UC大鼠有较好的疗效。其作用机理之一可能是通过增强机体抗氧化能力、抑制组织中的脂质过氧化反应,稳定细胞膜、从而保护肠粘膜、促进溃疡面愈合,达到治疗UC的作用。
[Abstract]:Objective: to observe the changes of disease activity index, colonic mucosa gross morphology and histopathology in UC model rats treated with Draconis and Houttuynia houttuyniae, and to detect the changes of serum superoxide dismutase (SOD) and malondialdehyde (MDA) (MDA) in UC model rats. To verify the therapeutic effect of Houttuynia cordata and Dragon Blood Draconis on UC, and to explore whether the combined use of Houttuynia cordata and Draconis has synergistic effect on UC and its mechanism, so as to provide the basis for the standardized treatment and medication of UC. Methods: SD rats were randomly divided into normal control group and model group. The model group was made by enema with 2: 4 and 6-trinitrobenzene sulfonic acid plus ethanol. When the UC model was successfully established, the rats in the model group were randomly divided into four groups: Dragon blood group, houttuynia group, low, middle and high dose group, (SASP) group and model control group. After two weeks of administration, the blood samples were collected from the heart to detect the serum levels of SOD and MDA, and the pathological colon was taken by laparotomy for pathological sections. Results: after intragastric administration, the body weight of rats in each gavage group was increased, and the body weight of rats in each group was higher than that in the middle dose group (P0.05), and there was no significant difference between the two groups (P0.05), while in the middle dose group and SASP group, there was no significant difference between them (P0.05), while in the middle dose group and SASP group, there was no significant difference between them (P0.05). Compared with Houttuynia cordata group, the weight gain was greater, the difference was statistically significant (P0.05). The hyperemia, edema, erosion and ulceration of colonic mucosa of rats in each group were improved, and there was no significant difference among the four groups (P0.05), and there was no significant difference among the four groups (P0.05), and there was no significant difference among the four groups (P 0.05), and there was no significant difference among the four groups, such as the combination of dragon blood and houttuynia houttuyniae. Compared with SASP group, the colon mucosal injury index of middle dose group, Dragon blood group and Houttuynia group was lower, the difference was statistically significant (P0.05). The concentration of SOD in serum was increased and the concentration of MDA in serum was decreased in each group. There was significant difference between middle dose group and SASP group, houttuynia group and dragon blood group in the range of Draconis houttuyniae and Houttuynia houttuyniae (P0.05). Conclusion: the experimental study shows that Dragon Blood Draconis combined with Houttuynia Houttuyniae is high. The middle dose group can improve the symptoms of congestion and edema, erosive bleeding, ulcer and other symptoms of colon mucosa, increase the activity of serum SOD and decrease the content of MDA, and it is superior to the SASP group, Houttuynia group and Dragon blood group. It was suggested that the high dose group had better curative effect on UC rats in the combination of Dragon Blood and Houttuynia cordata. One of its mechanisms may be to enhance the antioxidant ability of the body, inhibit lipid peroxidation in tissues, stabilize cell membrane, protect intestinal mucosa, promote ulcer healing, and achieve the effect of treating UC.
