辣椒素预防吲哚美辛致急性胃黏膜损伤的作用及机制

【摘要】:目的:研究不同时期辣椒素(CAP)对吲哚美辛致急性胃黏膜损伤的作用及机制。方法:成年雄性健康SD大鼠80只,随机分为8组,每组10只,实验分两期,Ⅰ期为2周,Ⅱ期为4周。Ⅰ期分别为A1组(空白对照组)、B1组(吲哚美辛模型组)、C1组(CAP预先给药组)、D1组(CAP预先给药+吲哚美辛模型组);Ⅱ期分组方法同I期。采用吲哚美辛复制大鼠胃黏膜损伤模型,于应激4 h后处死大鼠,收集胃液测定胃酸总酸度,肉眼计数胃黏膜损伤指数,行胃窦组织病理学观察,检测胃黏膜辣椒素受体(TRPV1)、降钙素基因相关肽(CGRP)的表达,检测胃窦组织丙二醛(MDA)、超氧化物歧化酶(SOD)及前列环素(PGI2)的水平。结果:I期:A1组、C1组胃黏膜未见损伤;与B1组相比,D1组胃黏膜损伤程度、胃酸总酸度、MDA值差异无统计学意义(P0.05),SOD、PGI2值显著升高(P0.05),TRPV1、CGRP表达显著增多(P0.05)。Ⅱ期:A2组、C2组胃黏膜未见损伤;与B2组相比,D2组胃黏膜损伤程度显著减轻(P0.05),胃酸总酸度显著降低(P0.05),MDA值显著降低(P0.05),SOD、PGI2值显著升高(P0.05),TRPV1、CGRP表达显著增多(P0.05)。结论:(1)CAP 1 mg/(kg·d)连续灌胃2、4周,对正常大鼠胃黏膜大体形态、病理组织学无损伤作用;(2)预先给予CAP 1 mg/(kg·d)连续灌胃4周具有预防吲哚美辛致大鼠急性胃黏膜损伤的作用。
