益生菌对溃疡性结肠炎大鼠肠黏膜TLRs表达的影响

【摘要】:目的:通过建立溃疡性结肠炎大鼠模型,探讨益生菌对溃疡性结肠炎(UC)大鼠模型肠黏膜Toll样受体(TLRs)表达的调控,及其在溃疡性结肠炎(UC)发生发展中可能的作用机制。方法:本实验分为正常对照组、模型组、金双歧治疗组、Toll样受体4单克隆抗体(TLR4mAb)干预组。采用三硝基苯磺酸(TNBs)法造模。对每组大鼠进行疾病活动指数(DAI)评分及组织病理学(HPS)评分,采用实时荧光定量(RT-PCR)法测定TLR4、TLR2mRNA的表达,酶联免疫吸附试验(ELISA)检测血清TNF-α、IL-1β的浓度。结果:模型组结肠组织TLR4、TLR2mRNA的表达及血清TNF-α、IL-1β的浓度均明显高于正常对照组,金双歧及TLR4mAb干预后,其表达均明显低于模型组,差异有统计学意义。结论:溃疡性结肠炎的发生与异常免疫反应有关。益生菌可改善溃疡性结肠炎大鼠的结肠炎症反应,其机制可能与影响肠黏膜TLRs表达及炎症因子的水平有关。
[Abstract]:Objective: to investigate the regulation of probiotics on the expression of Toll like receptor (TLRs) in intestinal mucosa of rats with ulcerative colitis (UC) by establishing a rat model of ulcerative colitis and its possible mechanism in the pathogenesis and development of ulcerative colitis. Methods: the experiment was divided into normal control group, model group and TLR4mAb intervention group. The model was made by (TNBs) method with trinitrobenzene sulfonic acid. The disease activity index (DAI) score and histopathological (HPS) score were measured in each group. The expression of TLR4,TLR2mRNA was measured by real-time fluorescence quantitative (RT-PCR) method, and the concentration of serum TNF- 伪 -1 尾 was detected by Elisa (ELISA). Results: the expression of TLR4,TLR2mRNA in colonic tissue and the concentration of serum TNF- 伪 -1 尾 in the model group were significantly higher than those in the normal control group. After the intervention of Jin Bifidobacterium and TLR4mAb, the expression levels were significantly lower in the model group than in the model group, and the difference was statistically significant. Conclusion: the occurrence of ulcerative colitis is related to abnormal immune response. Probiotics can improve the colitis response in rats with ulcerative colitis. The mechanism may be related to the influence of intestinal mucosal TLRs expression and inflammatory factor levels.
