重症急性胰腺炎大鼠回肠肌间神经节血管活性肠肽和一氧化氮合酶阳性神经元变化

【摘要】:目的:研究伴有胃肠动力障碍的重症急性胰腺炎(SAP)大鼠肠神经系统回肠肌间神经节血管活性肠肽(VIP)和一氧化氮合酶(NOS)阳性神经元的变化,以探讨SAP胃肠动力障碍的神经机制。方法:SD大鼠20只随机分为假手术组和SAP组,用5%牛黄胆酸钠逆行胰胆管穿刺注射制作SAP模型。24 h后测定小肠推进比及胰腺病理评分。制作回肠肌间神经节全层标本,应用双重免疫荧光染色,用荧光显微镜观察VIP和NOS阳性神经元的形态及计算其占总神经元的百分比。结果:与假手术组相比,SAP组大鼠腹腔腹水形成,肠系膜见钙化灶,肠管明显扩张,小肠推进比显著降低[(51.37±5.04)%vs.(30.20±3.82)%,t=10.570,P0.01],胰腺病理评分增高(3.74±0.23 vs.13.78±0.65,t=-45.858,P0.01),VIP和NOS阳性神经元的比例明显增高[(8.48±1.06)%vs.(14.22±2.12)%,t=7.642,P0.01;(29.04±3.36)%vs.(40.55±2.59)%,t=-8.570,P0.01]。结论 :SAP胃肠动力障碍可能与VIP和NOS两种抑制性神经元的表达上调相关。
[Abstract]:Aim: to study the changes of vasoactive intestinal peptide (VIP) and nitric oxide synthase (NOS) (NOS) positive neurons in ileal intermuscular ganglion (IGM) of severe acute pancreatitis (SAP) rats with gastrointestinal motility disorder (SAP) in order to explore the neural mechanism of SAP gastrointestinal motility disorder. Methods Twenty male Sprague-Dawley rats were randomly divided into sham operation group and SAP group. The SAP model was made by retrograde injection of 5% sodium taurocholate into the pancreatopancreatopancreatic duct. The small bowel propulsion ratio and pancreatic pathological score were measured after 24 hours.
